Abstract

In der vorliegenden Arbeit wird die Identifikation, Klonierung und Reinigung von acht homologen Holliday-Struktur auflösenden Endonukleasen (Hjcs) aus Archaeen und zwei homologen Hjcs aus archaealen Viren, sowie ihre spezifischen Reaktionen mit cruciformer DNA beschrieben. Die gereinigten Hjcs wurden in vergleichende Untersuchungen mit synthetischen cruciformen DNAs eingesetzt. Dabei zeigten alle Hjcs die erwartete, typische Auflösungsreaktion. Es konnte ein strukturabhängiger Schnittmodus abgeleitet werden, der sich zwischen den zellulären und den viralen Hjcs durch die Positionen der eingefügten symmetrischen Einzelstrangbrüche unterschied. Die zellulären Hjcs zeigten identische Schnittmuster mit einer cruciformen DNA mit fixiertem Kreuzungspunkt, unterschieden sich aber deutlich in ihren Reaktionen mit einer cruciformen DNA, die branch migration über einen Bereich von zehn Basenpaaren zuließ. Um Bereiche der Hjcs zu identifizieren, die für diese Spezifitäten verantwortlich sind, wurden drei hypothetische DNA-Interaktionsdomänen eines Hjcs einzeln und in Kombination in ein anderes Hjc mit stark abweichendem Schnittmuster übertragen. In Reaktionen mit mobiler cruciformer DNA zeigten die chimären Hjcs individuelle Schnittmuster, die sich von den Schnittmustern aller natürlicher Hjcs unterschieden. Dies zeigte den Einfluß der hypothetischen DNA-Interaktionsdomänen auf die Substratspezifität der Hjcs. Die archaealen Hjcs wurden in Experimenten zur Komplemention von Endonuklease VII des Bakteriophagen T4 eingesetzt. Die Fähigkeit der Hjcs zur Komplementation von Endo VII ließen Rückschlüsse auf die Funktion von Endo VII zu und bestätigten die biologische Funktion der Hjcs als X-Solvasen in einem in vivo-System.

This study describes the identification, cloning and purification of eight homologous Holliday-junction cleavage enzymes (Hjcs) from archaea and two homologous Hjcs from archaeal viruses, and their specific reactions with cruciform DNAs. The purified Hjcs were comparatively analysed in their reactions with synthetic cruciform DNAs. Here, all Hjcs showed the predicted activity typical for X-solvases. A structure dependent cleavage mode could be deduced for all Hjcs, which differed for the cellular and the viral Hjcs in the position of the symmetrical single strand breaks, introduced for resolving the Holliday-structure. The cellular Hjcs produced identical cleavage patterns in reactions with a cruciform DNA with a fixed junction, but showed different patterns in reactions with a cruciform DNA with a branch migratable junction. Certain regions of the Hjcs could be assigned to be potential DNA-interaction domains, being the structural basis of the observed specificities. To verify this hypothesis, chimeric Hjcs were constructed transferring the hypothetical DNA-interaction domains from one Hjc to another Hjc with a distinctively different cleavage pattern. In reactions with a mobile junction the chimeric Hjcs showed individual cleavage patterns, differing from all patterns produced by the natural Hjcs. Thus, an impact of the hypothetical DNA-interaction domains on the structure-specificity of the Hjcs could be confirmed. The archaeal Hjcs were used in assays to complement endonuclease VII of bacteriophage T4. The results suggest the impact of X-solvase activity of endonuclease VII's different functions for T4-development and provide direct evidence for the X-solvase activity of the Hjcs in an in vivo-system.English
