Abstract

Zusammenfassung Stämme des Gram-positiven Bakteriums Corynebacterium glutamicum stellen heutzutage die wichtigsten Organismen zur biotechnologischen Herstellung von Aminosäuren dar. Da C. glutamicum ein genetisch und physiologisch gut untersuchtes Bakterium ist und für dessen Manipulation gut funktionierende Transformationsmethoden und Vektorsysteme zur Verfügung stehen, gibt es Bestrebungen, auch Pantothensäure mit Hilfe dieses Organismus herzustellen. Der Weltjahresbedarf dieses ökonomisch wichtigen Vitamins wird heute durch chemische Verfahren gedeckt. Bei der Konstruktion eines Produktionsstammes für Pantothensäure sind nicht nur Manipulationen des Stoffwechsels, sondern auch der Transmembranflüsse von Produkt (Pantothenat) und Substrat (b-Alanin) wichtig, da diese Flüsse die Produktion ebenfalls beeinflussen können. Die am Transport von Pantothenat und b-Alanin beteiligten Systeme waren in C. glutamicum bisher noch nicht bekannt. Ziel dieser Arbeit war deshalb deren Charakterisation und Identifikation. Es konnte gezeigt werden, dass der Import von b-Alanin durch einen sekundär aktiven, Membranpotential-abhängigen Carrier mit einem Km-Wert von 5 mM und einer maximalen Geschwindigkeit Vmax von 1,66 nmol (mg TG)-1min-1 katalysiert wird. Die Identifikation des b-Alaninaufnahmesystems ist durch Homologiestudien nicht gelungen, jedoch konnte durch eine heterologe Expression des Gens für den b-Alanin Carrier aus Escherichia coli die b-Alaninaufnahmerate in C. glutamicum bis um das 16-fache gesteigert werden. Im Gegensatz zu b-Alanin wird Pantothenat durch ein primär aktives, ATP-abhängiges und hoch-affines System in die Zelle transportiert, welches sich durch einen Km-Wert von 0,9 µM und einer maximalen Geschwindigkeit von 26,6 pmol (mg TG)-1min-1 auszeichnet. Das Aufnahmesystem konnte durch Homologiestudien nicht gefunden werden, da ein ABC-Transportsystem für Pantothenat in Bakterien oder Säugern noch nicht bekannt ist. Für die Identifikation des Pantothenat-Exkretionssystems konnten in einem Screening von Transposonmutanten zwei Gene identifiziert werden, deren Deletion jeweils zu einem fast vollständigen Verlust der Pantothenatexkretion führte. Die Überexpression eines dieser Gene führte zu einer Verdoppelung des exkretierten Pantothenats.

Abstract Strains of the Gram-positive bacterium Corynebacterium glutamicum represent the most important organisms in the biotechnological production of amino acids. Since C. glutamicum is a genetically and physiologically well understood bacterium and established transformation methods and vector systems are available, there are efforts made to produce also pantothenic acid using this organism. The world's annual demand for this economically important vitamin is still met by chemical synthesis. Constructing a producer strain, one should not only take into account the possibilities of metabolic engeneering, but also the transmembrane fluxes of product (pantothenate) and substrate (b-alanine), which can be limiting processes. The systems involved in the transport of pantothenate and b-alanine are still not known in C. glutamicum. Therefore, the aim of this work was the characterization and identification of these systems. In the present work the uptake of b-alanine was found to be catalyzed by a secondary active, membrane potential-driven carrier which has a Km of 5 mM and a maximum velocity Vmax of 1,66 nmol (mg wd)-1min-1. The identification of the b-alanine uptake system was not possible by homology studies, but the heterologue expression of the b-alanine carrier CycA of Escherichia coli resulted in a 16-fold increase of the b-alanine uptake rate in C. glutamicum. The characterization of the pantothenate transport system revealed that C. glutamicum posseses a primary active, high-affinity carrier, which has a Km of 0,9 µM and a maximum velocity Vmax of 26,6 pmol (mg wd)-1min-1. Since an ABC transport system is still not identified in bacteria or mammals, the identification of the carrier by homology studies was not possible. For the identification of a pantothenate excretion system the screening of transposon mutants resulted in two genes, which were deleted individually. Both deletions led to an almost complete loss of pantothenate excretion, whereas an overexpression of one of the genes resulted in a twofold increase of pantothenate production.English
