Abstract

Die Fusion von "green fluorescent protein" (GFP) an Proteine ist eine geeignete Methode zur Untersuchung von dynamischen Prozessen in vivo. Die Grünalge Chlamydomonas reinhardtii ist ein Modellorganismus für die Analyse der eukaryotischen Geißel. In dieser Arbeit sollte die GFP-Technik für die Analyse von Proteinen des Basalapparates der Geißel untersucht werden. "Striated fiber assemblin" (SFA), Centrin und "deflagellation induced protein of 13 kDa" (DIP13), wurden N- (nur SFA) und C-terminal mit GFP fusioniert und stabil in C. reinhardtii transformiert. SFA ist Hauptbestandteil der quergestreiften Mikrotubuli-assoziierten Fasern (SMAFs). GFP-markiertes SFA wurde in diese Fasern eingebaut. N-terminal mit GFP fusioniertes SFA glich in seinen Eigenschaften dem Wildtypprotein und die Länge der Fasern nahm mit der Expressionsstärke zu. Die Kopfdomäne von SFA wird für die Faserbildung benötigt; "Photobleaching"-Experimente sprechen gegen eine ausgeprägte Dynamik der Fasern. Die C-terminale GFP Fusion von SFA besaß eine erhöhte Löslichkeit, und die Polymere waren instabil bei höheren Temperaturen. Die Analyse von Chlamydomonas Mutanten zeigte, dass die Anordnung der GFP-SFA Polymere von der Basalkörperanzahl und der Centrinmenge beeinflusst wird. Das Calcium-bindende Protein Centrin ist in den "nucleus basal body connectors" (NBBCs) und der Verbindungsfibrille zwischen den Basalkörpern lokalisiert. Hier wurde bei moderater Expression auch Centrin-GFP nachgewiesen. Überexpression führte zum Abbau der NBBCs, während die Verbindungsfibrille stabil war. In Induktionsversuchen wird das Fusionsprotein zunächst nur in die NBBCs eingebaut. Die Ergebnisse zeigten, dass Centrinfilamente unterschiedliche molekulare Eigenschaften besitzen. In vivo Beobachtungen zeigten weiterhin, dass die NBBCs elastisch sind. DIP13 ist homolog zum humanen Autoantigen NA14 und im Basalapparat lokalisiert; hier war auch DIP13-GFP zu finden. Allerdings führte die Überexpression dieses Fusionsproteins zu Aggregatbildung, Reduktion des Wildtypproteins und einem verändertem Phänotyp, der durch verlangsamtes Zell- und Geißelwachstum sowie dem Fehlen von Geißeln und erhöhter Letalität gekennzeichnet war. Obwohl die GFP-Markierung die Eigenschaften der untersuchten Proteine veränderte, konnten neue Informationen zur Funktion der Proteine und der aus ihnen gebildeten Strukturen gewonnen werden.

Fusion of green fluorescent protein (GFP) to proteins is a powerful method to investigate dynamic processes in vivo. The green flagellate Chlamydomonas reinhardtii is a model organism for studying the eukaryotic flagella. In this work the GFP-tagging of proteins was employed in order to analyse proteins of the flagellar basal apparatus. Striated fiber assembling (SFA), centrin and deflagellation induced protein of 13 kDa (DIP13) were tagged with GFP at the C-terminal domain. In addition SFA was tagged at the N-terminal domain. The chimeric genes were stably transformed in C. reinhardtii. SFA is the mayor component of the striated microtubule associated fibers (SMAFS). GFP tagged SFA was incorporated into this fibers. N-terminal tagged SFA had similar properties like the wild-type protein. The length of the fibers increased with the strength of expression. The head domain of SFA is essential for fiber formation and photobleaching experiments did not show a pronounced dynamic of the fibers. The C-terminally GFP-tagged SFA showed an increased solubility and the polymers were instable at higher temperatures. The correct number and arrangement of the fibers depended on the number of basal bodies and the amount of centrin as revealed by analysis of C. reinhardtii mutants. Centrin, a calcium binding protein, is localized in the nucleus basal body connectors (NBBCs) and the distal connecting fiber interconnects the two basal bodies. Moderate levels of expression allowed centrin-GFP to be incorporated into these structures. Over-expression of centrin-GFP disrupted the NBBCs whereas the connecting fiber was not affected. Shortly after induction of centrin-GFP the fusion-protein was observed at the NBBCs but was absent from the connecting fiber. In vivo observations using a strain with moderate centrin-GFP expression revealed flexible properties of the NBBCs. Wild-type DIP13, a homolog to human autoantigen NA14, as well as the GFP-fusion were located in the flagellar basal apparatus. However over-expression of DIP13-GFP resulted in aggregation of the protein, a reduced amount of wild-type protein and an altered phenotype. The cells had shorter flagella or even lacked flagella, cell growth was delayed, and lethality was increased. Although GFP-tagging alters the properties of the analysed proteins, this work gained new insights in the functions of the proteins and of the structures that are build out of these proteins.English
