Abstract

Diese Arbeit befasst sich mit der genetischen Charakterisierung des Gens LATERAL SUPPRESSOR (LAS) aus Arabidopsis thaliana und der Identifizierung neuer, an dem Prozess der Initiation von Lateralmeristemen beteiligter Gene. Der las-4-Mutante liegt eine 20 bp-Deletion im ersten Drittel des LAS-Gens zugrunde, die zu einer Unterdrückung der Anlage von Lateralmeristemen in den Achseln der Rosettenblätter führt. Im ersten Teil dieser Arbeit wurde die las-4-Mutante mit anderen Mutanten, die spezifische Entwicklungsdefekte zeigen, gekreuzt, um die Rolle von LAS bei der Initiation von Lateralmeristemen zu untersuchen. Eine Gruppe von Mutanten zeichnet sich durch ein verstärktes Auswachsen der Achseltriebe aus der Blattrosette aus. Die Analyse der axr1-12/las-4-, max1-1/las-4- und max2-1/las-4-Doppelmutanten zeigte in allen drei Fällen eine Unterdrückung der Seitensprossentwicklung aus den Achseln der Rosettenblätter. Somit wirkt las-4 in diesem Entwicklungsprozess epistatisch über axr1-12, max1-1 und max2-1. In der axr1-12/las-4-Doppelmutante wurde außerdem festgestellt, dass im Vergleich zur las-4 Mutante eine größere Anzahl der jüngeren Rosettenblätter Seitentriebe anlegte. Da in axr1-12-Mutanten verschiedene Mechanismen der Signaltransduktion des Phytohormons Auxin gestört sind, lässt dieses Ergebnis auf eine Rolle von Auxin sowohl bei der Initiation von Achselmeristemen als auch beim Auswachsen der Achselknospen schließen. Außerdem konnte im Rahmen der vorliegenden Studie eine Beteiligung von LAS bei der Initiation von Lateralmeristemen während der reproduktiven Phase nachgewiesen werden. So ist die Bildung akzessorischer Seitentriebe aus Stängelblättern von axr1-12/las-4-Doppelmutanten unterdrückt, ebenso unterdrückt las-4 bei der homöotischen Blüten-Mutante ap1-10 die Bildung ektopischer Blüten. Des Weiteren zeigten stm-2/las-4- und wus-1/las-4-Doppelmutanten synergistische Effekte in der Infloreszenz, was auf redundante Funktionen von LAS mit STM bzw. WUS bei der Entwicklung von Infloreszenzen hinweist. Außerdem deckte die Analyse von stm-2/las-4- und wus-1/las-4-Pflanzen eine gemeinsame Funktion bei der Trennung von Organen auf, da in beiden Fällen ausgeprägte Fusionen von Blattstielen festgestellt wurden. Das REVOLUTA-Gen ist wie LAS für die Anlage von Achselmeristemen nötig. Da in vorherigen Studien gezeigt werden konnte, dass rev die frühe postembryonale Organogenese von wus-1 und stm-2 teilweise unterdrückt, wurden wus-1/las-4 und stm-2/las-4-Pflanzen etabliert und charkterisiert. Die Analyse zeigte, dass las-4 die postembryonale Organogenese von stm-2 und wus-1 im Gegensatz zu rev nicht unterdrückt. Die Charakterisierung von rev-5/las-4-Doppelmutanten zeigte ausserdem, dass las-4 die Seitentriebe aus der Rosette, die rev-5 aufgrund seiner Variabilität des Phänotyps bildet, unterdrückt. Zusammen deutet dies darauf hin, dass beide Gene nicht im gleichen Signaltransduktionsweg fungieren. In einem zusätzlichen Experiment wurde ein Einfluss der Meristemidentität auf das Verzweigungsmuster von las-4-Mutanten untersucht. In las-4-Mutanten ist die Anlage von Lateralmeristemen in Achseln der vegetativen Rosettenblätter unterdrückt, nicht aber in den Achseln der Stängelblätter. Weil in lfy-14-Mutanten das SAM auch nach der Blühinduktion vegetativen Charakter behält, wurde die Mutation mit las-4 kombiniert. Zog man die Doppelmutanten unter Kurztagbedingungen an, war im Gegensatz zu las-4- und lfy-14-Einzelmutanten die Seitentriebbildung aus Achseln von Stängelblättern nahezu völlig unterdrückt. Daraus kann geschlossen werden, dass in las-4-Mutanten ein Impuls vom Infloreszenzmeristem zur Anlage von Achseltrieben aus Stängelblättern notwendig ist. In einem zweiten Ansatz wurde die las-4-Mutante chemisch mutagenisiert und unter den Nachkommen Mutanten identifiziert, die im Gegensatz zu las-4 keine Seitentriebe in Achseln von Stängelblättern anlegen. Es wurden 4 unabhängige, nicht allelische Linien, die enhancer of las-4 (eol) benannt wurden, genauer charakterisiert. Bei eol3/las-4-, eol5/las-4- und eol6/las-4-Pflanzen war die Anlage der Achselmeristeme aus Stängelblätter nur bei Anzucht unter Kurztagbedingungen unterdrückt, nicht aber unter Langtagbedingungen. eol1/las-4-Mutanten zeigten auch unter Langtagbedingungen eine reduzierte Verzweigung, waren aber spätblühend. Um einen der betroffenen Loci zu identifizieren, wurde zum Abschluss für eol3/las-4 eine Kartierung mit auf der PCR-Technik basierenden Markern durchgeführt. Die ermittelte Position befindet sich eng gekoppelt mit den Markern ciw12 und nga392 auf Chromosom1 und zeigt deshalb ebenfalls Kopplung mit dem LAS-Locus.

This study deals with the genetic characterisation of the LAS-gene of Arabidopsis thaliana and the identification of currently unknown loci involved in the process of lateral meristem initiation. The las-4 mutation, a 20 bp deletion towards the beginning of the LAS open reading frame, results in a suppression of lateral meristem initiation in axils of rosette leaves. The formation of side shoots from the axils of cauline leaves is not affected. In order to study the role of LAS in the process of lateral meristem initiation, the las-4 mutation was combined with other mutations that lead to specific developmental defects. A set of mutants is characterised by a enhanced outgrowth of lateral shoots from the rosette. Detailled analysis of axr1-12/las-4, max1-1/las-4 and max2-1/las-4 double mutants revealed a suppression of lateral meristem development from the axils of rosette leaves. This indicates that las-4 acts epistatic to axr1-12, max1-1 and max2-1 with respect to this process. Interestingly, in contrast to las-4 single mutants, in axr1-12/las-4 plants, the formation of lateral shoots was observed in a larger fraction from the oldest rosette leaves. As multiple aspects of auxin signalling are affected in axr1-12, this result suggests a role of auxin in the processes of both initiation and outgrowth of lateral shoots. Additionally, the suppression of the initiation of accessory shoots from axils of cauline leaves in axr1-12/las-4 plants indicates a role of LAS during reproductive development. This is supported by the observed suppression of ectopic flower formation from pedicels/peduncles in ap1-10/las-4 mutants. A redundant function of LAS with STM and WUS during development of inflorescences could also be concluded from the finding that both stm-2/las-4 and wus-1/las-4 mutants show synergistic effects in their inflorescences. The observation of severe fusions of petioles in both double mutants suggests an overlapping function of LAS with STM and WUS in organ separation. Like LAS, the REVOLUTA gene has been shown to be necessary for the initiation of lateral meristems. las-4 suppresses the variability of the rev-5 branching phenotype from the rosette. Taken together with the finding that las-4, in contrast to rev, does not suppress early postembryonic organogenesis in stm-2 and wus-1 mutants, this indicates that both genes act in separate developmental pathways. Because las-4 mutants fail to develop axillary meristems from axils of rosette leaves, but not from cauline leaves, the influence of meristem identity on the las-4 branching phenotype was analysed. A lfy-14/las-4 double mutant was established since the SAM of lfy-14 mutants maintains vegetative character even after floral induction. When grown under short day conditions, in these double mutants, the initiation of axillary meristems from cauline leaves was suppressed. This suggests an important influence of the reproductive meristem in axils of cauline leaves in las-4 mutants. In a second approach las-4 mutants were mutagenised and the progeny were screened for mutants with an enhanced branching phenotype compared to las-4. Four independent, non allelic enhancer of las-4 (eol) lines were characterised in detail. In eol3/las-4, eol5/las-4 and eol6/las-4 the initiation of lateral shoots from cauline leaves was suppressed only when the plants were grown under short day conditions. eol1/las-4 plants showed reduced branching under both long- and short day conditions and were late flowering. In a PCR based approach, the map position of EOL3 was determined. EOL3 is located on chromosome1, closely linked to markers ciw12 and nga 392 and is also linked to LAS.English
