Abstract

Matrix/Integrin-Interaktionen spielen eine wichtige Rolle bei verschiedenen physiologischen und pathophysiologischen Prozessen, wie der Embryonalentwicklung, Tumorinvasion und Wundheilung. Durch die Bindung von Kollagen Typ I an die Integrine a1b1 und a2b1 werden verschiedene zelluläre Effekte, wie die Induktion der Synthese von Matrix Metalloproteinasen (MMPs) und eine Reduktion der Kollagensynthese induziert. MMPs sind eine Familie von Zn2+-abhängigen Endopeptidasen die Bestandteile der extrazellulären Matrix degradieren können. Kollagen Typ I induziert im dreidimensionalen Kollagengel die Synthese von MMP-1 und MT1-MMP die Aktivierung von MMP-1 und MMP-2 in humanen dermalen Fibroblasten. Auch im Monolayer können ähnliche Effekte durch eine kollagen-imitierende Schlangengift-Metalloproteinase (SVMP), Jararhagin ausgelöst, die an den a2b1-Integrinrezeptor bindet. In dieser Arbeit wurde der Vorgang der Integrinaktivierung mittels verschiedener Domänen von integrinbindenden SVMPs analysiert und ein Zellkultursystem etabliert in dem Kollagenimitierende Effekte im Monolayer untersucht werden können. Zum einen wurde die Metalloproteinase-Domäne und eine darin enthaltende Integrinbindungsequenz (RKKH) genutzt, zum anderen die Disintegrinartige-Domäne und die darin enthaltende ECD-Sequenz, die an a2b1-Integrin binden und die Interaktion mit Kollagen Typ I inhibieren kann. Analysen der mRNA-Transkripte und Proteinmengen mittels Western Blot Analyse, Gelatine-Zymographien und Northern Blot-Analysen ergaben, dass beide Domänen die Synthese und Aktivierung von MMPs induzieren können. Dabei scheint die Disintegrinartige Domäne über die Bindung ihrer Integrinbindungssequenz ECD die MMP-Regulation zu beeinflussen. Die Wirkung der Metalloproteinase Domäne ist unabhängig von der RKKH-Sequenz und wird über die proteolytische Aktivität dieser Domäne vermittelt. Somit kann der Integrinrezeptor durch verschiedene Mechanismen aktiviert werden, um die MMP-Synthese und Aktivierung zu induzieren. Die Metalloproteinase Domäne kann auch andere zelluläre Effekte, die durch Kollagen-Integrin-Interaktionen ausgelöst werden, induzieren, wie eine reduzierte Kollagen Typ I Synthese und eine veränderte Zellmorphologie. Die Disintegrinartige Domäne kann diese Effekte nicht auslösen. Mittels der SVMPs und verschiedener Regionen oder Domänen aus diesen können verschiedene Effekte, die durch Kollagen induziert werden, imitiert und unabhängig von einander untersucht werden. Darüber hinaus wurde mit der Etablierung eines in vivo Modells zur Untersuchung der Funktion von MMPs begonnen, indem ein Mausstamm generiert werden soll, der eine gewebsspezifische MT1-MMP Defizienz in der Haut aufweist. Dazu wurde das Cre-loxP-System genutzt. Im Rahmen dieser Arbeit wurde der dazu benötigte Austauschvektor kloniert und die ersten stabilen Transfektionen von embryonalen Stammzellen mit diesem Vektor durchgeführt. In nachfolgenden Arbeiten sollen durch weitere Transfektionen embryonale Stammzellen generiert werden, die zur Etablierung transgener Mauslinien eingesetzt werden.

Interactions between integrins and the extracellular matrix (ECM) play a crucial role in several physiological and pathophysiological processes like embryonic development, tumour invasion and wound healing. The binding of integrins a1b1 and a2b1 to their ligand collagen type I induces a variety of cellular responses like reduced collagen type I synthesis and an increase in the synthesis of several matrix metalloproteinases (MMPs). MMPs are a family of Zn2+-dependent endopeptidases, that are able to degrade ECM components. Collagen type I induces the synthesis of MMP-1 and MT1-MMP and the activation of proMMP-1 and MMP-2 in fibroblasts grown in the three dimensional system of collagengels. Similar effects can be obtained in monolayer by the effect of a snake venom metalloproteinase (SVMP), jararhagin, that can bind to a2b1 integrin. In this study we analysed the activation of integrins by using distinct domains of two integrin binding SVMPs to establish a cell culture system, which can be used for investigations of collagen mimetic effects in monolayer. We used the metalloproteinase domain and a peptide (RKKH) corresponding to a described binding site in this domain. In addition we used the disintegrinlike domain and another peptide, containing the ECD Sequence of this domain, that have been shown to bind to a2b1 integrin and inhibit the interaction with collagen. Analysis of mRNA-transcripts and protein levels by using western blots, gelatine zymography and northern blots could show, that both domains can induce the synthesis and activation of the investigated MMPs. The disintegrinlike domain seems to work through its integrin binding on MMP-synthesis, whereas the influence of the metalloproteinase domain seems to be independent from a putative integrin binding. This domain can act through its proteolytic activity. The results indicate that integrins can be activated by different mechanisms to mediate MMP synthesis and activation. This finding is consistent with other results showing only the metalloproteinase domain to induce other integrin mediated cellular effects like reduced collagen type I synthesis and a change in cell morphology. The disintegrinlike domain has no effect. Incubation of Fibroblasts grown as monolayer with SVMP-domains is a useful in vitro cell culture system to investigate different collagen mimetic effects and to analyse these effects independently from one another. In addition we started to establish an in vivo model for further investigations of the role of MT1-MMP. Therefore we want to generate a conditional MT1-MMP deficient mousestrain by gene targeting using the Cre-loxP-System. During this study the targeting vector has been cloned and ES-cells have been successfully transfected with this vector. Further studies will continue with this project to generate a skin specific MT1-MMP deficient mouse strain.English
