Abstract

1. Im Rahmen dieser Arbeit wurden die native ISPS aus der Graupappel (Populus x canescens (Ait.) Sm.) und verschiedene heterolog in Escherichia coli exprimierte Derivate, mit C-terminalem bzw. N-terminalem His-Tag sowie unmarkiert, biochemisch charakterisiert. Die ISPS zeigte ein Temperaturoptimum von 40°C, ein breites pH-Optimum von pH 7 - 8,5 und einen apparenten Km-Wert im millimolaren Bereich. Zudem konnte festgestellt werden, dass die Enzymkinetik einen sigmoiden Verlauf aufwies, der auf eine positive Kooperativität des Enzyms hinweist. Die N-terminale Markierung der ISPS hat eine Verschiebung des pH-Optimums weiter in den alkalischen Bereich zur Folge, während die C-terminal markierte ISPS im Vergleich zum N-terminal markierten Enzym eine geringere Aktivität besaß. Durch die vergleichsweise hohe Affinität zum Substrat fiel jedoch der Unterschied bei geringen Substratkonzentrationen weniger stark aus. Das Temperaturoptimum ist bei C-terminaler Markierung um ca. 5-10°C erhöht, die Aktivierungsenergie um 50 %. Alle übrigen Eigenschaften blieben weitgehend konstant. 2. Zur Transformation von Pflanzen wurden zwei ispS-Genderivate hergestellt. Dazu wurde das ispS-Gen aus einer cDNA-Bank der Graupappel mit zwei unterschiedlichen Epitopen markiert, um das resultierende Protein besser nachweisen und aufreinigen zu können. Es wurde jeweils am C-terminalen Ende der ISPS ein Epitop angehängt, das HA-Epitop, ein Nonapeptid (YPYDVPDYA) und ein His-Tag, bestehend aus sechs aneinander gereihten Histidinen. 3. Die Funktionsfähigkeit der beiden ISPS-Derivate wurde durch heterologe Expression in E. coli überprüft. Dabei stellte sich heraus, dass das HA-Epitop zu einem völligen Verlust der Enzymaktivität der ISPS führte. Das Derivat mit His-Tag zeigte dagegen etwa 20 % der Aktivität der unverändert in Bakterien exprimierten ISPS. 4. Mit dem funktionsfähigen ISPS-Derivat mit His-Tag wurde Arabidopsis thaliana L. transformiert. Als Transformationsvektor wurde der binäre Vektor pBinAR verwendet, der einen 35S-Promotor und eine Kanamycin-Resistenz enthält. Die Übertragung auf die Pflanze erfolgte über Agrobacterium tumefaciens. 5. Es konnten 40 positive Linien über die Kanamycin-Resistenz selektiert werden, vier davon stammten aus einer ersten Transformation und konnten bereits geselbstet werden, die 36 aus einer späteren Transformation stammenden Linien lagen heterozygot vor. 6. Die stabile Insertion des ispS-Gens in das Genom von A. thaliana konnte für alle 40 Linien über PCR verifiziert werden. Die Genexpression konnte in 38 der 40 A. thaliana-Linien auf RNA-Ebene nachgewiesen werden. Eine erhöhte Isoprenemission konnte für fünf der 40 Linien gemessen werden, für die auch die Höhe der Genexpression über quantitative RT-PCR ermittelt wurde. Zwei der fünf Linien zeigten gleichzeitig zur erhöhten Isoprenemission eine erhöhte Genexpression. Als Grund für die geringe Erhöhung der Isoprenemission wird ein Substratmangel für die ISPS in A. thaliana vermutet, zu dem eine geringere Aktivität des Enzyms durch die C-terminale Markierung hinzukommt.

It is known that a lot of plant species emit high amounts of isoprene, especially during high temperature periods. The physiological impact of isoprene biosynthesis and emission is currently still unknown. An enhanced heat tolerance as well as an antioxidant action of isoprene is mainly discussed. One of the main goals of this work was therefore to produce transgenic plants differing from the corresponding wildtype in their ability to synthesize and emit isoprene. Therefore, the isoprene synthase (ispS) gene from poplar (Populus x canescens), which was isolated by MILLER ET AL. (2001) was used to transform Arabidopsis thaliana L., which is not a significant isoprene emitter. Prior to transformation the original DNA-sequence was extended by two different epitops, a nonapeptide HA epitope and six triplets for histidine resulting in a C-terminal His-tag, in order to get a labelled enzyme, which can be detected and cleaned up more easily afterwards. For proving the efficiency of the resulting proteins, the core enzymes without the transit peptide needed for the import of the protein, which is encoded in the nucleus, into the chloroplasts were expressed heterologous in E. coli. The HA epitope resulted in a complete loss of enzyme activity, while the His-tag led to a decreased enzyme activity of about 20 %. For the Agrobacterium mediated transformation of A. thaliana the ispS with the C-terminal His-tag was used and cloned into the binary vector pBinAR under the control of a 35S promoter. 40 transgenic lines, which were selected by kanamycine resistance, have been achieved. The stable integration of ispS was confirmed on DNA- as well as on RNA level. The expression of ispS was proved in 38 of the 40 lines by PCR from cDNA. Furthermore the emission of the transgenic lines was studied by measuring whole plants for several hours. Five of the 40 lines showed significant higher isoprene emission rates being more than 2,5 fold higher than in the measured non emitting A. thaliana plants. Two of the five new isoprene emitting plants showed coevally to the higher emission rates also a higher gene expression rate for the ispS, which was measured by quantitative RT PCR. In addition, native isoprene synthase (ISPS) from poplar as well as different derivates from it were characterised biochemically. The native enzyme and the unlabelled heterologous in E. coli expressed ISPS acted very similar. Isoprene synthase has a Km in the range of millimolar, a temperature optimum of 40 degrees and a broad pH-optimum of 7 to 8.5. Integration of an N-terminal His-tag led to a shifted pH-optimum to the alkaline side while the C-terminal His-tag had more influence. As indicated by the in vivo-expression analysis in E. coli, this manipulation led to a decrease in enzyme activity. The C-terminal his-tag also resulted in a higher temperature optimum and a higher activating energy.English
