Abstract

Mutationen im cartilage oligomeric matrix protein (COMP) führen beim Menschen zu Chondrodysplasien. In der vorliegenden Arbeit wurden zur Untersuchung der Pathogenese der Pseudoachondroplasie (PSACH) die Auswirkungen der am häufigsten identifizierten Mutation in COMP (del469D) sowohl im Zellkulturmodell als auch in transgenen Mäusen untersucht. Zur Analyse im Zellkulturmodell wurden sowohl primäre bovine als auch murine Knorpelzellen jeweils mit einem myc markierten Wildtyp bzw. mutiertem COMP Konstrukt transfiziert. Transgenes Wildtyp COMP wird von den Knorpelzellen kontinuierlich sekretiert und war extrazellulär mit einer kollagenhaltigen Matrix assoziiert. Mutiertes COMP zeigt dagegen eine deutlich verzögerte Sekretion mit eindeutiger Retention im ER. Der trotz Mutation sekretierte COMP Anteil war nicht mit der Kollagenmatrix assoziiert, sondern löslich im Überstand nachweisbar. Außerdem war die Vitalität in Zellen, die mutiertes COMP exprimieren, beeinträchtigt. In einem zweiten Ansatz wurden transgene Mäuse hergestellt, die Wildtyp und mutiertes COMP knorpelspezifisch exprimieren. Die Expression des Transgens wurde eindeutig auf RNA Ebene und Proteinebene nachgewiesen und quantifiziert. Transgene und endogene COMP Untereinheiten bilden gemischte, pentamere Moleküle, unabhängig davon, ob das transgene COMP mutiert ist. Mutiertes COMP ist schwerer aus Knorpelgewebe extrahierbar als Wildtyp COMP, was auf eine veränderte Verankerung in der ECM hinweist. Transgenes COMP konnte durch IF Färbung auf Paraffinschnitten in der Proliferationszone der Tibia Epiphyse nachgewiesen werden. Dabei zeigt Wildtyp COMP eine gleichmäßige Verteilung in der gesamten ECM, wohingegen mutiertes COMP auf die perizelluläre Matrix begrenzt war. EM Analysen zeigten sowohl mangelhaft ausgeprägte Kollagenfibrillen im perizellulärem Bereich als auch ungeordnete Zellcluster in der Proliferationszone. Im Sternum konnte eine Fusion des dritten und vierten Segments festgestellt werden, die eindeutig auf die Expression von mutiertem COMP zurückzuführen war, und auf einen Einfluss von COMP in der endochondralen Ossifikation deutet. Männliche Tiere zeigten einen Trend zu einem geringeren Längenwachstum. Sowohl radiologische Analysen als auch die Untersuchungen verschiedener Knochenparameter weiblicher Tiere mittels pQCT zeigten keine signifikanten Unterschiede zu Kontrollmäusen. Der eher schwache skeletale Phänotyp ist möglicherweise auf eine zu hohe endogene COMP Menge zurückzuführen, weshalb die Mäuse mit COMP defizienten Tieren gekreuzt wurden. Auch wenn der skeletale Phänotyp im Vergleich zum Patienten eher schwach ausfiel, erlaubt die Maus erste Einblicke in molekulare Mechanismen der Pathogenese von PSACH in vivo.

Mutations in the cartilage oligomeric matrix protein (COMP) cause chondrodysplasias in humans. The effects of the most common mutation in COMP (del469D) on the pathogenesis of pseudoachondroplasia (PSACH) were analysed in two experimental systems. In a cell culture model, primary bovine and murine chondrocytes were transfected with myc tagged wild type and mutant COMP variants. Transgenic wild type COMP was continuously secreted and furthermore integrated into a collagenous extracellular matrix. In contrast, secretion of mutant COMP was delayed due to a distinct retention within the ER. A small amount of mutant COMP still secreted dit not associate with the extracellular matrix (ECM). In addition, the viability of cells expressing mutant COMP was decreased. As a second system, transgenic mouse lines with a cartilage specific expression of wild type or mutant COMP were generated. The transgene expression was confirmed on the RNA and quantified on the protein level. Transgenic and endogenous COMP monomers were able to form mixed, pentameric molecules. In comparison with wild type COMP, mutant COMP was more difficult to extract from cartilage tissue indicating an altered anchorage within the ECM. Transgenic COMP was detected in the proliferative zone of the tibia epiphysis by immunofluorescence staining. Wild type COMP showed a uniform distribution within the ECM while mutant molecules were restricted to the pericellular matrix. Analysis by EM demonstrated improperly formed collagen fibrils in this region as well as disorganized cell clusters within the proliferative zone. Furthermore, transgenic mice show intersternebral fusion of the third and fourth sternebrae as a result of the expression of mutant COMP, indicating a role for COMP during endochondral ossification. Male mice exhibit a tendency for growth retardation, while radiological analysis as well as measurement bone parameter by pQCT revealed no significant differences between mutant and control mice. Even though the skeletal phenotype was milder than expected with regard to the clinical features seen in PSACH patients, the transgenic mouse model allows novel insights into in molecular mechanisms involved in the pathogenesis of PSACH.English
