Abstract

In der vorliegenden Arbeit wurden die Mechanismen der Apoptoseexekution durch selektive COX-2-Blockade mit dem NSAR SC-58635 in den HCC-Zelllinien HepG2 und HuH-7 untersucht. Während der Apoptoseexekution wurde die frühzeitige Aktivierung der Initiator-Caspase-8 und der Effektor-Caspasen CASP-3 und CASP-9 demonstriert und funktionell bestätigt. Weiter ergab die selektive COX-2-Blockade eine rasche Aktivierung des mitochondrialen Apoptosesignalweges. SC-58635 führte in den Mitochondrien zu einer schnellen Herunterregulierung von MCL-1, ein antiapoptotisches BCL-2-Familienmitglied, gefolgt von der Translokation von BAX und BID an die Mitochondrien und der CYT-C-Freisetzung aus den Mitochondrien ins Zytoplasma. Diese Vorgänge waren mit der Depolarisation des mitochondrialen Membranpotentials verbunden. In der Konsequenz führte die Überexpression von MCL-1 zu einer deutlichen Abnahme und die Blockade von endogenem MCL-1, mittels siRNA-mcl-1, zu einer Verstärkung der SC-58635 induzierten Apoptose. Die Beteiligung des extrinisischen Signalweges wurde zunächst über die Reduktion der Apoptose nach Überexpression von FADD dom. neg., einer mutanten Form des FADD-Proteins, demonstriert. Zudem wurde durch die selektive COX-2-Blockade die Oberflächenexpression der Todesrezeptoren CD95, TNF-R1, TRAIL-R1 und TRAIL-R2 reguliert und die Rezeptorsysteme für ihre entsprechenden Liganden sensitiviert. Eine Ausnahme hiervon bildete das CD95-Rezeptorsystem von HuH-7-Zellen mit mutiertem p53. Weiterhin beeinflussten FADD, CASP-8 und CASP-3 auch die Depolarisierung der Mitochondrienmembran, womit ein Einfluss des extrinsischen Signalweges auf den intrinsischen Signalweg deutlich wurde. Von therapeutischer Relevanz war der Synergieeffekt von SC-58635 mit den chemotherapeutischen Wirkstoffen Bleomycin und Doxorubicin bei der Apoptoseinduktion sowie die Verstärkung der Apoptose in Kombination mit Mitoxantron in den untersuchten Zelllinien. Da dies unabhängig davon, ob p53 als WT oder in der mutierten Form vorlag, geschah, stellt die Kombination mit SC-58635 einen Vorteil gegenüber der klassischen chemotherapeutischen Behandlung, welche zumeist auf WTp53 angewiesen ist, dar. Weiterhin sensitivierte SC-58635 PHHs nicht für die chemotherapeutisch induzierte Apoptose, was die klinische Relevanz dieser Ergebnisse bestätigt.

In the present work the mechanisms by which Cyclooxygenase (COX)-2 inhibition through the NSAR SC-58635 induces apoptosis were investigated in the hepatocellular carcinoma (HCC) cell lines HepG2 containing WTp53 and HuH-7 possessing a mutated transcriptional inactive form of p53. The early activation of the initiator-caspase CASP-8 and the effector-caspases CASP-3 and CASP-9 during the apoptosis execution was demonstrated and functionally confirmed. The findings showed a link between selective COX-2 inhibition and the mitochondrial apoptosis pathway. SC-58635 led to a rapid down-regulation of myeloid cell leukaemia-1 (MCL-1), an anti-apoptotic member of the BCL-2 family, followed by translocation of BAX and BID to mitochondria, CYT-C release from mitochondria and depolarisation of the mitochondrial membrane potential. Consequently, overexpression of MCL-1 led to the inhibition of SC-58635 mediated apoptosis. In addition, blocking endogenous MCL-1 function using a small-interfering (si) RNA approach enhanced COX-2 inhibitor-mediated apoptosis. Furthermore, overexpression of a dominant negative FAS associated death domain mutant (FADD dom. neg.) reduced COX-2 inhibitor-mediated apoptosis, which demonstrated the participation of the extrinsic pathway. Additionally, SC-58635 triggered cell surface expression of the CD95, TNF R and TRAIL-R1 and -R2 death receptors rapidly and sensitized the investigated receptorsystems for their spcific ligands. An exception of this was the CD95 receptor in HuH-7 cells with mutated p53. Additionally, FADD dom. neg. as well as the inhibitors of CASP-8 and CASP-3 also reduced the depolarisation of the mitochondrial membranes. In order to that the extrinsic pathway via the death receptors showed an influence on the mitochondrial intrinsic pathway. It is of clinical importance that SC-58635 acted synergistically with the chemotherapeutic drugs bleomycin and doxorubicin in the induction of apoptosis and it amplified apoptosis in combination with mitoxantrone. This was independent of the WT or mutated form of p53 in the investigated HCC cell lines, which is an advantage over the dependency of WTp53 during classical chemotherapeutic treatment. The clinical relevance of these results is further substantiated by the finding that SC-58635 did not sensitize primary hepatocytes (PHH) toward chemotherapy-induced apoptosis.English
