Abstract

Adenylat bildende Enzyme nehmen eine zentrale Funktion in vielen Stoffwechselwegen ein. Sie katalysieren die Reaktion von Carbonsäuren mit ATP zu einem AMP-Anhydrid (dem Adenylat) und den darauf folgenden Transfer an einen Acyl-Endakzeptor, oftmals Coenzym A. In dieser Arbeit wurde eine Gruppe von Proteinen analysiert, die auf Grund ihrer Sequenz- und Strukturähnlichkeiten zu den 4-Cumarat:CoA-Ligasen (4CL) als putative Adenylat bildende Enzyme identifiziert, aber größtenteils noch nicht charakterisiert wurden. Diese Proteine werden als 4CL-ähnliche Proteine (4CLL) bezeichnet. Zur Charakterisierung der 4CLLs wurden diese zunächst heterolog exprimiert, aufgereinigt und einem umfangreichen Substratscreen mit rund 100 Carbonsäuren aus verschiedenen strukturellen Klassen unterzogen. Dabei zeigte sich, dass die Mehrheit der untersuchten 4CLLs als Acyl-CoA-Synthetasen von Fettsäuren verschiedenster Kettenlängen und Sättigungsgrade fungiert. Dies konnte durch einen in vivo-Assay bestätigt werden, welcher auf der Komplementation eines Acyl-CoA-Synthetase-defizienten E. coli-Stammes basiert. Somit aktivieren sechs der neun überprüften 4CLLs Fettsäuren und bilden damit, neben den bereits bekannten Acyl-CoA-Synthetasen langkettiger Fettsäuren (LACS), eine weitere Gruppe von Acyl-CoA-Synthetasen in A. thaliana. Zusätzlich aktivieren einige 4CLLs Derivate und Vorstufen der Phytohormone Jasmonsäure (JA) und Indolessigsäure (IAA). Das Enzym At1g20510 konnte als OPC-8:CoA-Ligase identifiziert werden. Es aktiviert eine Zwischenstufe der JA-Biosynthese, OPC-8, zum korrespondierenden CoA-Ester. Diese Funktion konnte in vivo durch Analysen von Verlustmutanten des entsprechenden Gens, die reduzierte Mengen an JA aufwiesen, bestätigt werden. Mit At4g05160 und At5g63380 konnten erstmals Enzyme beschrieben werden, die das IAA-Derivat Indolbuttersäure (IBA) in den korrespondierenden CoA-Ester überführen. Neben der unter in vitro-Bedingungen geringen, aber signifikanten IBA:CoA-Ligase-Aktivität zeigte die Analyse von Doppelmutanten beider Gene deutlich veränderte IAA- und IBA-Mengen. Des Weiteren konnte At1g62940 als Acyl-CoA-Synthetase von Hydroxyfettsäuren identifiziert werden und ist vermutlich an der Biosynthese von Sporopollenin in Blütenknospen beteiligt. Die funktionale Analyse der 4CLLs wurde jeweils durch Untersuchungen zur Expression der korrespondierenden Gene ergänzt. Insgesamt ergibt sich damit ein umfassendes Bild einer Gruppe von CoA-Ligasen, deren Vertreter differenzierte, wichtige Funktionen im Stoffwechsel von A. thaliana übernehmen.

Adenylate-forming enzymes are of fundamental importance in many metabolic pathways. These enzymes catalyze the reaction of a carboxylic acid with ATP to an AMP-anhydrid (the adenylate) and its subsequent transfer to an acyl acceptor, which in most cases is coenzyme A. In this study, the function of a largely uncharacterized group of proteins from Arabidopsis thaliana was systematically explored. Based on their high sequence and structural similarity to 4-coumarate:CoA ligase (4CL) these enzymes were named 4CL-like (4CLL) proteins and are thought to display adenylate-forming activity. To identify enzymatic functions, the heterologously expressed and purified 4CLL enzymes were subjected to an extensive substrate screen using about 100 carboxylic acids from different structural classes. These studies revealed that the majority of the tested 4CLLs function as acyl-CoA synthetases in vitro, activating fatty acids of variable chain length to the corresponding CoA esters. This function was confirmed by an independent in vivo assay, which relies on functional complementation of an acyl-CoA synthetase-deficient E. coli strain. In fact, six out of nine tested 4CLL enzymes showed this activity, thereby defining a new group of fatty acyl-CoA synthetases in A. thaliana, which is distinct from the known long-chain fatty acyl-CoA synthetase (LACS) family. In addition, several 4CLL enzymes were also active on various derivatives of phytohormones such as jasmonic acid (JA) and indoleacetic acid (IAA). Importantly, the enzyme At1g20510 efficiently activated OPC-8 to the CoA ester, the predicted intermediate of JA biosynthesis, thus representing an OPC-8:CoA ligase. This in vitro function was confirmed by analysis of various A. thaliana lines deficient for the corresponding gene, which showed reduced levels of JA. By contrast, the enzymes At4g05160 und At5g63380 showed week but significant in vitro activity towards indolebutyric acid (IBA) and in the corresponding A. thaliana mutant line the IAA/IBA balance was impaired. These results demonstrate that both enzymes participate in auxin metabolism and identify for the first time IBA:CoA ligases. Finally, the enzyme At1g62940 was found to activate hydroxy fatty acids, which are important precursors of sporopollenin biosynthesis in flowers of A. thaliana. The functional analysis of all enzymes was complemented by expression studies of the corresponding genes. Collectively, a comprehensive picture of a functionally important class of enzymes is presented.English
