Abstract

Das Nesprin-2 Giant Protein (796 kDa) ist ein Mitglied der α-Aktinin-Superfamilie und zeichnet sich durch das Vorhandensein einer Aktin-Binde-Domäne (ABD-Domäne),einer Spektrinwiederholungen enthaltenden Stabdomäne und einer C-terminalen Transmembrandomäne aus. Mittels des C-Terminus wird Nesprin-2 Giant in der inneren und äußeren Kernmembran verankert. Im Rahmen dieser Arbeit wurde die strukturelle Funktion des Nesprin-2 Giant Proteins bei der Aufrechterhaltung der Zellkernmorphologie untersucht. Hierzu wurden primäre dermale Fibroblasten einer Laminopathie-Patientin analysiert, die eine einzigartige Mutation im N-terminalen Bereich des Lamin A/C Proteins aufweist (LMNA S143F). Die Patientin zeigt einen kombinierten Phänotyp, der Charakteristika einer Progerieerkrankung und Myopathie vereint. Einhergehend mit der bekannten Abhängigkeit der Nesprine von Lamin A/C kann in diesen Zellen eine veränderte Expression und Lokalisation von Nesprin-2 Isoformen, insbesondere Nesprin-2 Giant, beobachtet werden. Um zu zeigen, dass die beobachteten Veränderungen eindeutig auf die fehlende Funktion des Nesprin-2 Proteins zurückzuführen sind, wurden Nesprin-2 Giant defiziente Mäuse analysiert. In LMNA S143F Zellen sowie in Nesprin-2 Giant defizienten Fibroblasten und Hautschnitten geht der Verlust des Nesprin-2 Giant Proteins mit Kerndeformationen und einer Vergrößerung der nukleären Fläche einher. Es konnte gezeigt werden, dass das Expressionsniveau des Nesprin-2 Giant Proteins wichtig für das Ausmaß der Kerndeformationen ist. Mittels Immunpräzipitation und RNA silencing wurde nachgewiesen, dass die Lokalisation der kleinen C-terminalen Isoformen an der Kernmembran von der Anwesenheit des Nesprin-2 Giant Proteins abhängig ist. Neben strukturellen Veränderungen der Zellkernmorphologie konnte in der Nesprin-2 Giant defizienten Maus ein Einfluss auf Proteine der LEM-Domäne-Familie (Emerin, LAP2α, LAP2β) sowie Veränderungen in einem chromatin-assoziierten Protein nachgewiesen werden. Diese Veränderungen sind typische Kennzeichen von Zellen mit LMNA Mutationen und wurden bislang als direkte Konsequenz von Lam A/C Mutationen angesehen. In dieser Arbeit konnte erstmals nachgewiesen werden, dass Nesprine einen direkten Einfluss auf Proteine nehmen, die eine wichtige Rolle bei der Genregulation und Chromatinorganisation spielen. Somit konnte gezeigt werden, dass das Nesprin-2 Giant Protein eine potentielle Rolle in der Pathogenese von Laminopathien einnimmt, indem es Einfluss auf die strukturelle und funktionelle Integrität des Zellkerns ausübt.

Nesprin-2 Giant (796 kDa) is a member of the α-actinin-superfamiliy and consists of an actin-binding-domain (ABD-domain), a rod domain containing several spectrin repeats and a C-terminal KASH domain which mediates anchorage to the outer and inner nuclear membrane. In order to characterise the function of this protein, primary cells and tissues obtained from Nesprin-2 Giant deficient mice as well as primary dermal fibroblasts of a patient carrying a LMNA S143F mutation in the N-terminal segment of Lamin A/C were analysed. The patient shows a unique phenotype combining features of progeria and myopathy. Consistent with the known Lamin A/C dependency of Nesprin-2, expression levels and localisation of Nesprin-2 isoforms, predominantly Nesprin-2 Giant, were perturbed. In LMNA S143F fibroblasts as well as primary cells and skin sections of Nesprin-2 Giant deficient mice, nuclear deformations were frequently observed and accompanied by an increase in nuclear size demonstrating that the level of Nesprin-2 Giant expression is crucial for the induction of nuclear defomations. Using immune precipitation analysis and RNAi silencing a Nesprin-2 Giant dependency of the smaller c-terminal Nesprin-2 isoforms was confirmed. Besides the structural changes, an aberrant expression and localisation of LEM-domain-containing proteins (Emerin, LAP2α, LAP2β) as well as a mislocalisation of the heterochromatin associated protein HP1β could be observed in Nesprin-2 Giant deficient mice. As these changes are hallmarks of fibroblasts obtained from patient with laminopathies they were considered as a direct consequence of LMNA mutations. Here we show that Nesprins directly influence proteins important for gene regulation and chromatin organisation. We can conclude that Nesprins play an essential role in the pathogenesis of laminopathies by influencing the structural and functional integrity of the nucleus.English
