Abstract

Die Kartoffelpflanze ist eine der wichtigsten Kulturpflanzen der Welt. Ihre Knollen enthalten hauptsächlich Stärke, die bis zu 80 % des Trockengewichts ausmachen kann. Zusätzlich zur Nahrungsmittelproduktion liefert sie Rohstoffe für industrielle Applikationen. So war und ist es nach wie vor ein Ziel der klassischen Pflanzenzüchtung sowie der biotechnologischen Forschung, den Stärkeertrag pro Pflanze zu erhöhen. Ein Bestandteil der vorliegenden Arbeit waren Untersuchungen von biotechnologisch veränderten Kartoffelpflanzen, welche die ATase2 aus Arabidopsis thaliana unter Kontrolle des 35S-CaMV- (Cauliflower Mosaic Virus) oder des B33-Patatin- Promotors überexprimierten. Einen weiteren Bestandteil stellte die Analyse von Freilandanzuchten bereits beschriebenen (Zhang et al., 2008) transgener Kartoffelpflanzen mit erhöhter GPT- und NTT-Aktivität in ihren Knollen dar. Beide Ansätze verfolgten das Ziel, Erhöhungen im Stärkegehalt und/oder im Ertrag von Kartoffelknollen pro Pflanze zu bewirken. Eine Überexpression der ATase2 unter Kontrolle des 35S-Promotors konnte im Vergleich zum WT lediglich tendenziell erhöhte Stärkegehalte in Kartoffelknollen und Blättern bewirken. Es traten keine signifikanten Änderungen im Knollenertrag pro Pflanze auf. Überexpression der ATase2 unter Kontrolle des B33-Promotos hatte ebenfalls nur tendenziell erhöhte Stärkegehalte von Knollen zur Folge, was jedoch zu Lasten des Knollenertrags pro Pflanze ging. Kartoffelpflanzen mit erhöhter GPT- und NTT-Aktivität in ihren Knollen zeigten bei Anzuchten im Gewächshaus signifikante Steigerungen im Stärkegehalt und im Ertrag von Kartoffelknollen pro Pflanze im Vergleich zum WT. Bei Freilandanzuchten wiesen die transgenen Pflanzen jedoch eine leichte Verringerung im Stärkegehalt und im Ertrag von Kartoffelknollen pro Pflanze auf. Darüber hinaus zeigten sie insgesamt schlechteres Wachstum der oberirdischen Pflanzenteile und früher einsetzende Seneszenz. Die Ursachen der Unterschiede zwischen Gewächshaus- und Freilandanzuchten konnte nicht abschließend geklärt werden.

Potato plants are one of the most important crops in the world. Potato tubers mainly contain starch, which constitutes up to 80 % of their dry weight. In addition to food production potato plants deliver resources for industrial applications. So it has ever been an aim of classical breeding and biotechnological research to improve the starch content and yield of potato tubers. One part of this treatise was the analysis of biotechnologically modified potato plants which overexpressed the ATase2 from Arabidopsis thaliana under control of the 35S-CaMV- (Cauliflower Mosaic Virus) or of the B33-Patatin-promotor. Another part was the analysis of field trials with potato plants exhibiting an increased GPT- and NTT-activity in their tubers which had already been described (Zhang et al., 2008). Both approaches had the ambition to increase the starch content and/or the tuber yield per plant. Overexpression of ATase2 under control of the 35S-promotor merely caused trends to result in higher starch contents in tubers and leaves. Significant changes in tuber yield per plant did not emerge. Overexpression of ATase2 under control of the B33-promotor caused trends to result in higher tuber starch contents as well, at the expense of tuber yield per plant, however. Potato plants with increased GPT- and NTT-activity in tubers grown in the greenhouse exhibited significant increases in tuber starch contents and tuber yield in comparison with the WT. In case field trials, however, the starch content and the yield of tubers per plant were slightly reduced. Moreover they exhibited a worse growth of the aerial parts of the plant and an earlier senescence. The reasons for these differences between greenhouse and field trials could not be finally clarified.English
