Abstract

Die kleine Rho-GTPase Rac1 besitzt wichtige Funktionen in der Regulation des Aktinzytoskeletts, der Proliferation und der Migration von epithelialen Zellen. Aufgrund von Experimenten mit Knockout-Mäusen und transgenen Mäusen wird Rac1 eine wichtige Rolle in der Erhaltung des epidermalen Stammzellpools sowie in der Wundheilung der Haut zugeschrieben. In den bisher untersuchten Mäusen mit Epidermis-spezifischer Deletion von Rac1 konnten keine differenzierten Untersuchungen zu spezifischen Funktionen von Rac1 in verschiedenen Kompartimenten der Epidermis und ihrer Anhangsgebilde durchgeführt werden, da die Deletion immer im germinativen Kompartiment erfolgte und daher alle daraus hervorgehenden Zellen Rac1 negativ waren. Durch Einkreuzung eines mutierten Rac1 als Transgen in den Knockout-Hintergrund haben wir sogenannte Rac1 Rescue Mäuse erzeugt, in denen die Rac1-Aktivität selektiv im Keratin 14-positiven Kompartiment der Epidermis und der Haarfollikel rekonstitutiert wurde. Rac1 Rescue Mäuse zeigen im Gegensatz zum epidermalen Rac1 Knockout ein Wachstum der Haare sowie erkennbare Haarfollikel. Die infolge des Verlusts von Rac1 beobachteten Störungen der Haarmorphogenese können durch basale Rekonstitution der Rac1-Aktivität vollständig aufgehoben werden. Jedoch sind die Haarfollikel von Rac1 Rescue Mäusen im Vergleich zu Kontrolltieren verkürzt. Ursache hierfür ist offenbar eine verminderte Proliferation von Keratinozyten der Haarmatrix. Weiterhin sind Rac1 Rescue Mäuse durch ein mattes, gräulich erscheinendes Fell gekennzeichnet. Dies geht einher mit einer veränderten Haaroberflächenstruktur und einer gestörten Pigmentverteilung im Haarschaft. Mit dieser gestörten Pigmentverteilung korrelieren Defekte in der regulären Anordnung der Zellen der Haarmedulla sowie eine gestörte Ausbildung von Ausstülpungen des Haarcortex, die für die Adhäsion zwischen Cortex und Medulla verantwortlich sind. Die im Rahmen dieser Arbeit erhobenen Daten lassen damit erstmals auf eine spezifische Funktion von Rac1 in der Strukturbildung und Pigmentierung der Haare schließen. Eine Suche nach Ursachen für die Haarfollikeldegradierung nach Rac1-Deletion führte zu dem Ergebnis, dass diese durch die Induktion von Apoptose in der äußeren Haarwurzelscheide des Haarfollikels mitverursacht sein könnte. Weitergehende funktionelle Untersuchungen zeigten, dass die Apoptose-Resistenz von epidermalen Keratinozyten mit Rac1-Defizienz oder mit gehemmter Rac1-Funktion nach UV-Bestrahlung stark vermindert ist. Dies ist die erste Evidenz für eine direkte Funktion von Rac1 in der Regulation der UV-Licht-induzierten Apoptose. Die molekularen Mechanismen der Beeinflussung der Apoptose-Resistenz durch Rac1 sollen den Gegenstand weiterer Untersuchungen bilden.

The small Rho-GTPase Rac1 has important functions in the regulation of the actin cytoskeleton as well as in the proliferation and migration of epithelial cells. Experiments with knockout and Rac1 transgenic mice have shown its important role in the maintenance of the epidermal stem cell pool and in skin wound healing. In the mice with epidermis specific deletion of Rac1 described so far, detailed investigation of specific functions of Rac1 in the different compartments of the epidermis and its appendages could not be performed. Due to the deletion in the germinative compartment all progeny were negative for Rac1. By introducing a mutated Rac1 as a transgene into the knockout background we have generated mice in which Rac1 activity has been selectively restored in the Keratin 14-positive compartment of the epidermis and hair follicles. These mice are referred to as Rac1 Rescue mice. In contrast to epidermis specific Rac1 knockout mice, Rac1 Rescue mice show intact hair follicles and growth of hair. The observed retardation in hair morphogenesis caused by the loss of Rac1 can be restored completely by the basal reconstitution of Rac1 activity. However, hair follicles of Rac1 Rescue mice are shorter than those of control mice apparently due to reduced proliferation of the hair matrix keratinocytes. Moreover, the coat of Rac1 Rescue mice is dull and grey instead of black. This appearance of the coat correlates with a disturbed pigment distribution and surface structure of the hair shaft. In addition, we have detected defects in the regular arrangement of the medulla cells as well as an impaired formation of hair cortex protrusions responsible for the adhesion between cortex and medulla. The data shown here suggest, for the first time, a specific function of Rac1 in the formation and pigmentation of hairs. When searching for possible reasons for the hair follicle degradation in Rac1 E-KO mice we observed an increased frequency of apoptotic keratinocytes in the outer root sheath of the hair follicles. Further functional investigations have demonstrated that resistance to apoptotic cell death of epidermal keratinocytes deficient for Rac1 or of keratinocytes with inhibited Rac1 function is markedly reduced upon UV irradiation. This is the first evidence for a direct function of Rac1 in the regulation of UV-induced apoptosis. The molecular mechanisms by which Rac1 regulates the apoptotic resistance of keratinocytes will be subject of further studies.English
