Abstract

Die regulatorische PP2A Untereinheit phr2AB ist das D. discoideum Ortholog von B55, der regulatorischen Untereinheit in Säugern. Sie wurde als Bindepartner des zentrosomalen Proteins CEP161 identifiziert. Zielsetzung dieser Arbeit ist, die Interaktion mit unabhängigen Methoden zu bestätigen und zu untersuchen, ob diese Interaktion direkt oder indirekt ist. Darüberhinaus soll herausgearbeitet werden, an welchen Bereich von CEP161 phr2AB bindet. In Immunfluorezenzanalysen soll die Lokalisation von phr2AB festgestellt werden. Schließlich sollen die Konsequenzen einer Überexpression von phr2AB als GFP-Fusionsprotein auf Eigenschaften von D. discoideum analysiert werden.

The regulatory subunit phr2AB in Dictyostelium discoideum is the ortholog of the mammalian regulatory subunit B55. Phr2AB was identified as the binding partner of the centrosomal protein CEP161. Aim of this work is to confirm the interaction between phr2AB and CEP161, as well as investigate which part of CEP161 is binding phr2AB. Another aspect of this work is the consequences of an overexpression of GFP-tagged phr2AB on the characteristics of D.discoideum. Furthermore, the localisation of phr2AB will be investigated with immunofluorescence studies.
