The glycolipids and phosphatidylglycerin of the chloroplast, formed in vivo and in vitro, result from a de novo synthesis of the uniform diglyceride component sn-1-oleoyl-2-palmitoylglycerin. Transformed into the monoene lipid, the species that represent structural and functional components in the membranes of the chloroplasts are formed. As mass spectroscopic analyses and positional analyses of lipids from isolated envelopes show, these species are
{C-1}/{C-2} {18:3}/{16:3}-MGD, {18:3}/{16:0}-DGD,{18:3}/{16:0}-SQD,
{18:3}/{3-tr-16:1}-PG.
Lipid-coupled desaturation is discussed as responsible for the production of these lipid-specific fatty acid patterns. C18 desaturases could convert the oleic acid of all lipids mentioned, but C16 desaturases are specific for MGD. In the synthesis of fatty acid combinations of hexa species
{C-1}/{C-2} {18:3}/{18:3}-MGD, {18:3}/{18:3}-DGD,
the desaturation and elongation of palmitic acid in relation to the C-2 position of the MGD plays a central role.
A significant contribution of phosphatidylcholine components to galactolipid synthesis in “C16:3 plants” is therefore excluded. This confirms glycolipid synthesis by purified, physiologically intact chloroplasts. | English |
