Abstract

Das Arylcyclohexylamin Ketamin hat aufgrund seiner außergewöhnlich schnell einsetzenden und nachhaltigen antidepressiven Wirkung bei bislang therapieresistenten Patienten ein starkes weltweites Interesse geweckt. Die Depressionsbehandlung mit Ketamin wird allerdings durch dessen dissoziative Nebenwirkungen und Missbrauchspotenzial sowie einer schlechten oralen Bioverfügbarkeit limitiert. Der Ketamin-Metabolit Hydroxynorketamin zeigte dahingegen in Nagetierversuchen einen vergleichbar schnellen antidepressiven Effekt, ohne Ketamin-typische Nebenwirkungen oder ein Abhängigkeitspotenzial zu generieren. Der Hauptteil der vorliegenden Arbeit beschäftigt sich mit der Synthese von Derivaten von Ketamin-Metaboliten. Neben der Synthese Ketamin-analoger Verbindungen über verschiedene literaturbekannte Verfahren, deren Substratspektrum sich als begrenzt erwies, wurde ein neues synthetisches Verfahren, über das die Kernstruktur von Ketamin-Derivaten mit Hilfe einer Diels-Alder-Reaktion konstruiert wird, entwickelt. Die Grundstruktur Ketamin-artiger Verbindungen, welche sich über literaturbekannte Methoden nicht herstellen ließen, wurde über die in dieser Arbeit entwickelte Synthese aufgebaut, was deren synthetischen Nutzen verdeutlicht. Die hergestellten Derivate von Ketamin-Metaboliten wurden am Deutschen Zentrum für Luft- und Raumfahrt (DLR) in Köln in vitro an primären hippocampalen murinen Neuronen auf deren pro-neuroplastische Aktivität untersucht. In den Zellexperimenten wurde eine im Vergleich zu Ketamin und Hydroxynorketamin wesentlich stärkere Stimulation der Bildung präsynaptischer Boutons beobachtet, was mit einem hohen neuroregenerativen und antidepressiven Wirkpotenzial korreliert.

The arylcyclohexylamine ketamine has generated a strong worldwide interest due to its extraordinary rapid-acting and sustained antidepressant effect in previously treatment-resistant patients. However, depression treatment with ketamine is limited by its dissociative side effects and abuse potential, as well as its poor oral bioavailability. The ketamine metabolite hydroxynorketamine, on the other hand, showed a comparably rapid antidepressant effect in rodent experiments, without generating ketamine-typical side effects or a dependence potential. The main part of the present work describes the synthesis of derivatives of ketamine metabolites. The synthesis of ketamine-related compounds via publicized methods proved a low substrate range. Hence a new synthetic procedure with a Diels-Alder reaction for the construction of the core structure of ketamine derivatives was developed. The basic structural motifs of ketamine-related compounds which were not accessible via literature-know methods, were build up with the new developed synthesis, illustrating its synthetic utility. The pro-neuroplastic activity of the synthesized derivatives of ketamine metabolites was studied in vitro on primary hippocampal murine neurons at Deutsches Zentrum für Luft- und Raumfahrt (DLR) in Cologne. In the cell experiments a significant greater stimulation of the formation of presynaptic boutons was observed compared to ketamine and hydroxynorketamine. The strong increase of the presynaptic boutons correlates with a high neuroregenerative and antidepressant action potential.UNSPECIFIED
