Abstract

Die intensive Nutzung von Antibiotika hat zur raschen Verbreitung von antibiotikaresistenten Pathogenen geführt. Es besteht deshalb der dringende Bedarf an neuen Antibiotika, die keine Kreuzresistenz mit anderen Substanzklassen zeigen. Das Dipeptid-Antibiotikum TAN1057 zeigt in Tiermodellen sowie unter geeigneten Testbedingungen in vitro exzellente antibakterielle Wirkung gegen Staphylokokken einschließlich S. epidermidis und Methicillin-resistenten S. aureus Stämmen (MRSA). Im Rahmen dieser Arbeit wurden der Wirkmechanismus von TAN1057 und die molekularen Ursachen der Resistenzentwicklung in S. aureus untersucht. In mehreren unabhängigen Experimenten stieg unter Selektionsdruck über 6 Tage die Resistenz der Bakterien täglich um den Faktor 2 bis 4 an. Nach sechstägiger Selektion lagen hochresistente S. aureus Stämme mit stabiler Resistenz (MHK: >64 µg/ml) vor. Die TAN1057-resistenten S. aureus Stämme wiesen keine Kreuzresistenz gegenüber anderen antibakteriellen Substanzen auf. Dieser Befund unterstützt die Hypothese, dass TAN1057 die bakterielle Translation mittels eines neuen Wirkmechanismus inhibiert. Mittels eines in vitro Transkriptions-/Translationssystems wurde gezeigt, dass sich die TAN1057-Resistenz auf ribosomaler Ebene in einem einzelnen Sprung während der sechstägigen Resistenzselektion manifestierte. Neben der resistenzvermittelnden Modifikation des Wirkortes von TAN1057 tragen demnach auch andere Ursachen, wie z. B. eine verringerte Substanzaufnahme oder ein Efflux-Mechanismus zur Gesamtresistenz bei. Zur Analyse der ribosomalen Resistenz wurden PCR-Primer entwickelt, die erstmalig die selektive Amplifikation und Sequenzierung aller 23S-rRNA Operons, der Pseudouridinsynthasen sowie sämtlicher L-Proteine in S. aureus ermöglichten. Die Selektion TAN1057-resistenter S. aureus Stämme führte reproduzierbar zu Bakterienstämmen einer spezifischen Glycindeletion innerhalb des ribosomalen L2-Proteins. Diese Mutation wurde vor dem Anstieg der ribosomalen Resistenz nachgewiesen und konnte demnach nicht der Auslöser der ribosomalen Resistenz sein. Aufgrund des reproduzierbaren, TAN1057-spezifischen Auftretens während der Resistenzselektion sowohl in S. aureus als auch in E. coli besteht zwischen der Glycindeletion im ribosomalen Protein L2 und dem Wirk- bzw. Resistenzmechanismus von TAN1057 jedoch ein ursächlicher Zusammenhang.

The extensive use of antibiotics has led to the rapid spread of drug resistant pathogens. There is a critical need for new antimicrobial compounds lacking cross resistance to other substance classes. The dipeptide antibiotic TAN1057 displays excellent antibiotic activity in vivo and in vitro under suited test conditions against staphylococci, including S. epidermidis and methicillin resistant S. aureus (MRSA) strains. In this work, we investigated the mode of action of TAN1057 and the molecular basis of resistance development in S. aureus against this compound. Independant resistance selection experiments over a period of six days revealed a daily increase of bacterial resistance by a factor of 2 to 4. After six days, highly resistant S. aureus strains with stable MICs against TAN1057 (>64 µg/ml) were observed. The selected TAN1057 resistant S. aureus strains displayed no cross-resistance against other antimicrobial compounds. This result supports the hypothesis that TAN1057 inhibits bacterial translation by a new mode of action. Coupled in vitro transcription/translation experiments revealed that the TAN1057 resistence emerged on a ribosomal basis as a single step during the six day resistance selection. A target modification that leads to an increase in TAN1057 resistance is therefore accompanied by other causes of bacterial resistance, like lower uptake of the compound or increased cellular efflux. To analyse the ribosomal resistance mechanism in detail, PCR-primers were designed for specific amplification and sequencing of the chromosomal operons for 23S-rRNA, pseudouridine-synthases and all ribosomal L-proteins in S. aureus. The selection of TAN1057 resistant S. aureus strains reproducibly yielded bacteria encoding a mutant ribosomal protein L2 with one specific glycine deletion. The TAN1057 specific mutation was observed before the increase of ribosomal resistance, indicating that this mutation is not the direct elicitor for the increase in ribosomal resistance. The reproducible, TAN1057 specific appearance of this mutation during the resistance selection in S. aureus and E. coli indicates nonetheless a causal connection of the observed glycine deletion in protein L2 during the TAN1057 resistance selection and the mode of action of TAN1057.English
