Abstract

Die vorliegende Arbeit beschreibt die Synthese, strukturelle Charakterisierung und Untersuchung der cytotoxischen Eigenschaften einer systematischen Reihe von Platin(II)- und Palladium(II)-Komplexen mit zweizähnigen Phosphan-Liganden der Art cis-[MXY(R2P-(CH2)n-PR2)] (mit M = Pt, Pd; X, Y = Cl, C6F4R' (R' = F, OMe, OEt, OnPr); R = Ph, Et, Me; n = 1-5). Als Syntheseroute zur Darstellung der gewünschten Verbindungen diente die mit hohen Ausbeuten ablaufende Decarboxylierungsreaktion. Eine Modifikation der Reaktion durch Austausch des toxischen Thallium(I)-polyfluorbenzoates gegen ungiftiges Kalium-polyfluorbenzoat war möglich und bewährte sich sowohl bei der Synthese von Palladium(II)-Komplexen als auch jener von Platin(II)-Komplexen mit fragilen Liganden. Die Kristallstrukturen der meisten synthetisierten Produkte konnten aufgeklärt werden. Dabei handelte es sich in fast allen Fällen um Komplexe des gewünschten Typs. Eine Ausnahme bildeten jedoch die beiden verbrückten, zweikernigen Komplexe trans-[PtCl(C6F5)-{mu-(dpppe)}2-PtCl(C6F5)] und trans-[PtCl(C6F5)-{mu-(dpppe)}2-Pt(C6F5)2]. Cytotoxische Eigenschaften konnten für alle untersuchten einfach polyfluorphenylsubstituierten Komplexe sowohl in sensiblen L1210- als auch Cisplatin-resistenten murinen L1210/DDP-Leukämie-Zellkulturen nachgewiesen werden. Der Komplex cis-[PdCl(C6F5)(dppp)] erwies sich als äußerst potentes Cytostaticum, dessen Wirkung auf sensiblen L1210-Zellkulturen (IC50 0,875 mumol/l) der des Cisplatin (IC50 0,5 mumol/l) sehr nahe kommt und die des Cisplatin in Cisplatin-resistenten L1210/DDP-Zellkulturen um das Zehnfache übersteigt. Der Vergleich der IC50-Konzentrationen zeigte eine Abhängigkeit der Cytotoxizität der einfachsubstituierten Polyfluorphenyl-Komplexe des Platin(II) und Palladium(II) mit zweizähnigen Phosphan-Liganden sowohl von der Länge der verbrückenden Kette des Phosphan-Liganden, als auch dessen Substituenten am Phosphor sowie der Art der Polyfluorphenyl-Gruppe und des Metalles auf. Demnach sollte im Falle von cis-[MXY(R2P-(CH2)n-PR2)] (M = Pd, X = Cl, Y = C6F4OMe, R = Alkyl, n = 2) eine maximale cytotoxische Aktivität vorliegen. Des Weiteren scheint eine Korrelation zwischen den kristallographisch bestimmten Metall-Chlor-Abständen und den IC50-Werten der Verbindungen zu bestehen, aus der sich für cis-[PdCl(C6F5)(dmpe)], welches bislang noch nicht auf seine Cytotoxizität hin untersucht wurde, sehr gute zellteilungshemmende Eigenschaften herleiten lassen. Dies würde wiederum mit den zuvor abgeleiteten optimierenden Parametern einhergehen. Des Weiteren lässt die Abhängigkeit der Cytotoxizität von dem Metall-Chlor-Abstand auf eine Involvierung des labilisierten Chlor-Liganden in den Wirkungsmechanismus schließen.

The present work describes the synthesis, structural characterization and investigation of the cytotoxic activities of a systematic series of platinum(II) and palladium(II) diphosphine complexes of the type cis-[MXY(R2P-(CH2)n-PR2)] (where M = Pt, Pd; X, Y = Cl, C6F4R' (R' = F, OMe, OEt, OnPr); R = Ph, Et, Me; n = 1-5). The compounds have been synthesized by the decarboxylation reaction. A modification of the decarboxylation reaction by replacing the toxic thallous(I) polyfluorobenzoate by harmless potassium polyfluorobenzoate was successful and revealed a good reaction pathway for the synthesis of palladium(II) complexes and platinum(II) complexes with fragile ligands. The crystal structures of most of the compounds were determined and exhibit monomeric molecular structures with exception of the bridged, binuclear complexes trans-[PtCl(C6F5)-{mu-(dpppe)}2-PtCl(C6F5)] and trans-[PtCl(C6F5)-{mu-(dpppe)}2-Pt(C6F5)2]. Cytotoxic activity could be detected for all investigated monopolyfluorophenyl substituted complexes in sensitive L1210 and cisplatinum-resistant L1210/DDP mouse leukemia cell lines. The complex cis-[PdCl(C6F5)(dppp)] exhibited the highest cytotoxic activity (IC50 0,88 mumol/l) which is comparable to that of cisplatinum (IC50 0,5 mumol/l) in sensitive L1210 cell lines and an even 10-fold higher activity (IC50 0,65 mumol/l) in cisplatinum-resistant L1210/DDP cell lines (IC50 6,9 mumol/l). The results of the antitumor activity investigations suggest an influence of the phosphorus substituents, the bridging carbon chain, the polyfluorophenyl group and the metal towards the cytotoxicity of the monopolyfluorophenyl substituted compounds. Following these results the complex cis-[MXY(R2P-(CH2)n-PR2)] (M = Pd, X = Cl, Y = C6F4OMe, R = Alkyl, n = 2) should reveal the highest antitumor activity. Additionally, there appears to be a correlation between the M-Cl bond length (M = Pd, Pt) in the crystal structure to the cytotoxic activity. This correlation indicates that cis-[PdCl(C6F5)(dmpe)] which has not been examined yet should be the most active complex synthesized as part of this thesis. This result agrees with the interpretation of the influence of the phosphorus substituents, the bridging carbon chain, the polyfluorophenyl group and the metal. Additionally, the correlation between the M-Cl bond length and cytotoxicity points to a reaction mechanism in which the labilized chlorine is involved.English
