Abstract

Die dorsoventrale Musterbildung wird beim Langkeim-Insekt Drosophila melanogaster durch lokalisierte Toll-Rezeptor Aktivierung induziert, was zur Ausbildung eines Kernkonzentrationsgradienten des rel/NF-kB Proteins Dorsal führt. Zwei Zielgene von Dorsal sind twist und snail. Sie werden auf der ventralen Seite des Embryos aktiviert, wo die Kernkonzentration von Dorsal am höchsten ist. Um herauszufinden, ob beim Kurzkeim-Insekt Tribolium castaneum ein ähnlicher Mechanismus in der dorsoventralen Achsenbildung verwendet wird, wurden die Gene Toll, twist und snail studiert. Puppale RNA-interference (RNAi) mit Toll verursachte den Verlust der dorsoventralen Polarität, da die Embryonen für zen und dpp symmetrische Expressionsmuster zeigten. Zudem exprimierten die Toll-RNAi Embryonen kein twist im Rumpfbereich. Ein wichtiger Unterschied zu Drosophila zeigt sich im Expressionsmuster von TcToll: in späten Oozyten und Präblastoderm Embryonen sind keine Toll-Transkripte detektierbar. Allerdings wird Toll während der Keimstreifaustreckung sehr stark in den Kopflappen exprimiert, was vermuten lässt, dass es in Tribolium anders reguliert wird als in Drosophila. RNAi mit twist, einem wichtigen mesodermalen Gen, verursacht den Verlust der Ventralfuchenausbildung und der Expression von snail. Anders als bei Drosophila führt der Verlust von twist nicht zu einer gravierenden Störung der larvalen Kutikula. Die snail-RNAi Phänotypen zeigten keine Unterschiede zu den twist RNAi Phänotypen, da snail von twist reguliert wird. Diese Beobachtungen deuten darauf hin, dass grundlegende Schritte in der dorsoventralen Musterbildung zwischen Drosophila und Tribolium konserviert sind.

In the long-germ insect Drosophila melanogaster dorsoventral polarity is induced by localized Toll-receptor activation which leads to the formation of the nuclear gradient of the rel/NF-kB protein Dorsal. Two targets of Dorsal are twist and snail, that are activated at most ventral regions where the highest concentrations of the nuclear Dorsal protein are found. To investigate whether the short-germ insect Tribolium castaneum uses a similar mechanism of axis formation as Drosophila, we were studying the Toll, twist and snail genes from Tribolium. Pupal RNA-interference (RNAi) with Toll causes the loss of the dorsoventral polarity resulting in dorsalized embryos which show symmetrical patterns of zen and dpp expression and lack twist expression in the trunk region. A major difference to Drosophila is observed in the expression pattern of TcToll: no transkripts are detectable in late oocytes and preblastodermal embryos. However, a Toll expression-domain in the headlobe detectable until late germband-extension stages suggests differences in the regulation in Tribolium. RNAi with twist, an important mesodermal gene in Drosophila, abolishes ventral furrow formation and snail expression. There are only minor cuticular defects, but most of the larvae are not able to hatch probably because of muscle defects. The phenotypes observed with snail dsRNA injections differ not from the twist dsRNAi phenotypes, since snail is a target gene of twist. These observations indicate that fundemantal steps in the formation of the dorsoventral axis are conserved between the long germ Drosophila and the short germ Tribolium embryos.English
