Abstract

Corynebacterium glutamicum akkumuliert als Reaktion auf osmotischen Stress kompatible Solute. Hierzu synthetisiert das Bakterium die Aminosäuren Glutamat, Glutamin und Prolin de novo und/oder importiert die Solute Betain, Ectoin und Prolin mittels der sekundären Transporter BetP, EctP, LcoP und ProP, die jeweils unterschiedliche Affinitäten und Substratspektren zeigen. Die beteiligten Systeme der Osmoregulation werden auf der Ebene der Aktivität reguliert. Die Expressionsregulation der Transporter war bisher unbekannt. In dieser Arbeit wurde die Bedeutung der Expressionskontrolle der Transportergene betP, ectP, lcoP und proP sowie die Neusynthese kompatibler Solute nach einem hyperosmotischen Schock untersucht. Hierzu wurden der Wildtyp und Deletionsmutanten, die jeweils nur noch ein intaktes Gen eines Transporters für kompatible Solute hatten, auf Expressionsebene verglichen. Die Induktion der jeweiligen Gene wurde durch RNA-Hybridisierungsexperimente bestimmt und eine Quantifizierung der Transkriptionsstärke über Transportmessungen und Antikörpernachweise durchgeführt. Durch die Analyse von Wachstumsparametern und der Akkumulation von de novo synthetisierten Aminosäuren (Glutamat, Glutamin und Prolin) wurden zusätzliche wichtige Faktoren der Osmoregulation von C. glutamicum untersucht. Es konnte gezeigt werden, dass die Gene betP, ectP, lcoP und proP einer osmotischen Kontrolle unterliegen. Unter niederosmolaren Bedingungen wurden nur geringe Mengen an mRNA nachgewiesen, aber bereits eine hohe Grundaktivität der Transporter BetP und EctP festgestellt. Durch hyperosmotischen Schock wurden insbesondere die Gene betP, ectP und proP deutlich induziert. Die Transportraten von BetP und ProP stiegen aufgrund der dadurch verstärkten Translation signifikant an, während keine Steigerung der Rate bei EctP festgestellt werden konnte. Unter den getesteten Bedingungen zeigte lcoP nur eine schwache Reaktion, was darauf hindeutet, dass der Transporter in der Osmoadaptation eine geringe Rolle spielt. Aufgrund der hohen basalen Transportraten unter niederosmolaren Bedingungen bilden EctP und BetP somit das Notfallsystem von C. glutamicum bei plötzlich auftretenden Änderungen der Osmolalität und externer Verfügbarkeit kompatibler Solute. Zusätzlich wurde eine starke transiente Akkumulation von Glutamat sowie Glutamin nachgewiesen, welche in der frühen Osmoantwort zur schnellen Restaurierung des sinkenden Turgordrucks dienen und später durch hohe Konzentrationen von Prolin ersetzt werden, welches bis in die stationäre Phase das dominante Solut darstellt. Durch diese Untersuchungen konnte gezeigt werden, dass C. glutamicum über ein System verfügt, welches unter hyperosmotischen Bedingungen durch kumulative Nutzung verschiedener zellulärer Prozesse die zur Kompensation von osmotischem Stress notwendige Akkumulation kompatibler Solute gewährleistet.

To counteract the effects of hyperosmotic stress on turgor and cellular processes Corynebacterium glutamicum accumulates high amounts of compatible solutes. Therefore, the bacterium synthesizes the amino acids glutamate, glutamine, and proline de novo and/or imports the solutes betaine, ectoine, and proline via the secondary transporters BetP, EctP, LcoP, and ProP. These carriers exhibit different substrate affinities and spectra. The systems involved in osmoregulation are regulated at the level of activity. However, no data were available so far about the regulation of the transporters at the level of expression. This thesis analyzes the impact of expression control on the transporter genes betP, ectP, lcoP, and proP after hyperosmotic upshock. The C. glutamicum wild type strain and mutant strains devoid of four of the five genes each for transporters of compatible solutes were analyzed in detail. Induction of the respective genes was determined by RNA hybridization experiments. Quantification of expression intensity was carried out by transport measurements and antibody detection experiments. Analyses of growth parameters and determination of intracellular amounts of amino acids synthesized de novo (i.e. glutamate, glutamine, and proline, respectively) provided additional information on osmoregulation in C. glutamicum. It could be shown that the genes betP, ectP, lcoP, and proP are subject to osmotic control. Conditions of weak osmotic strength accounted for only small amounts of the respective mRNA's, though a surprisingly high basic activity for BetP and EctP was determined. Following osmotic upshock the genes betP, ectP, and proP were dramatically induced and the uptake rates of BetP and ProP rose significantly according to increased translation. However, no increase in the uptake rate of EctP was determined. LcoP showed only weak response under the tested conditions, which may be a hint to its minor role in osmoadaptation. Thus, BetP and EctP represent the emergency system of C. glutamicum when it is subject to a rapid increase of osmolarity and if external compatible solutes are available. Nevertheless, a transient accumulation of high concentrations of glutamate and glutamine could be determined in the first response to osmotic upshock. These amino acids were able to restore the decreasing turgor until being replaced by high concentrations of proline, which was the dominant compatible solute until stationary phase was reached. This study reveals the fine-tuned osmoprotective system of C. glutamicum which ensures the life-saving accumulation of compatible solutes by the use of multiple cellular processes under hyperosmotic conditions.English
