Abstract

Die Architektur einer Pflanze wird im Wesentlichen durch die Anordnung der Blätter und die Anzahl und den Grad des Auswachsens ihrer Seitentriebe bestimmt. In den Achseln der Blattprimordien werden neue Meristeme, so genannte Lateralmeristeme, angelegt. Diese Meristeme entwickeln sich zu Seitentriebknospen, die später zu Seitentrieben auswachsen. In der lateral suppressor- 4-Mutante (las-4) aus Arabidopsis ist in den Achseln von Rosettenblättern die Initiation von Lateralmeristemen unterdrückt, während in der reproduktiven Phase Seitentriebe in den Achseln der Stängelblätter angelegt werden (Greb et al., 2003). Die ls-Mutante aus Tomaten kann mit einem genomischen Fragment, welches das offene Leseraster und regulatorische Elemente des Arabidopsis LAS-Gens umfasst, komplementiert werden (Greb et al., 2003). Eine Mutation im orthologen Gen MONOCULM1 aus Reis führt zu einer Unterdrückung der Anlage von vegetativen Trieben (Li et al., 2003). Die LAS-Funktion ist also zwischen monokotylen und dikotylen Pflanzen konserviert. Ziel der Arbeit war die Charakterisierung regulatorischer Bereiche des LATERAL SUPPRESSOR Gens aus A. thaliana. Mit Hilfe von Deletionskonstrukten konnte der Promotor des LAS-Gens näher definiert werden. Eine Komplementation des las-4- Seitentrieb-Phänotyps ist mit einem Konstrukt, dass stromaufwärts mindestens 820 nt und stromabwärts einen 3'-Bereich von 4346 nt besitzt, möglich. Ein um 20 Nukleotide auf 800 nt verkürztes Konstrukt verliert diese Fähigkeit. In ersten in vitro Protein-DNA-Interaktionsstudien sind mit der Nukeotidsequenz von -790 bis -810 ein MYB-Transkriptionsfaktor (MYB22; At4g34990) und mit der Nukleotidsequenz von -820 bis -800 zwei zur Familie der TCP-Gene gehörende Transkriptionsfaktoren (TCP6; At5g23280 und TCP22; At5g08070) als mögliche Regulatoren von LAS gefunden worden. Ein weiteres notwendiges aber nicht ausreichendes regulatorisches Element befindet sich im Bereich -2910 bis -1447 vor dem Translationsstart. Ein im 3'-Bereich liegendes Element zwischen +3133 und +3547 kann diese Funktion ersetzen. Dies ist in Übereinstimmung mit Ergebnissen aus Tomate, die zeigten, dass eine Komplementation durch ein Konstrukt mit einem 5'-Bereich von 1392 nt und einem 3'- Bereich von 2623 nt möglich ist (Schmitt, 1999). Promotoraustausch-Experimente zeigten, dass das 3'-Tomaten-Element funktional in Arabidopsis ist. Dies lässt die Vermutung zu, dass beide 3'-Bereiche gleiche Module besitzen. Die Insertion einer T-DNA (-696; SALK 50989) und die damit verbundene Deletion führt zu einem Verlust von einem oder mehreren Modulen im 5'-Bereich zwischen -696 und -265. Eine Suche nach weiteren Bindemotiven zeigte innerhalb des 5'-Bereiches in Arabidopsis und des 3'-Tomaten-Elements Bindestellen für MADS-Box-Proteine, so genannte CArGBoxen. GUS-Analysen der jointless;lateral suppressor- Doppelmutante lassen vermuten, dass JOINTLESS die Position der Abszissionszone bestimmt. Ein im Vergleich zum Wildtyp schwächeres Signal in der ls-Mutante deutet auf eine autoregulatorische Transkription von Ls hin. Ein weiteres Ziel der Arbeit war die Isolierung von LATERAL SUPPRESSOR homologen Genen aus Gerste. Zwei in der Literatur beschriebene Gersten- Mutanten (uniculm1 und uniculm2) weisen starke Defekte in ihrer Seitentriebbildung auf (Kirby, 1973). Kartierungsanalysen und Sequenzierung der Kandidatengene in den verschiedenen Gersten- Mutanten zeigten, dass die Phänotypen der uniculm1- und uniculm2-Mutanten nicht auf einen Defekt im HvLs1- bzw. HvLs2-Gen zurückzuführen sind.

The architecture of plants is determined by the number, arrangement and growth intensity of their sideshoots. Sideshoots are initiated by the formation of axillary meristems in the axils of leaves. The las-4 mutant of Arabidopsis fails to initiate axillary meristems during vegetative development, whereas in the reproductive phase sideshoots are formed in the axils of cauline leaves (Greb et al., 2003). It is possible to complement the tomato mutant with a 6.3 kb genomic fragment, harbouring the Arabidopsis LAS ORF, as well as -2.9 kb of 5'-sequence and 2.1 kb of 3'-sequence (Greb et al., 2003). A mutation in the orthologes gene from rice MONOCULM1 (MOC1) leads to a suppression of tiller formation (Li et al., 2003). The aim of this work was to identify and characterize upstream regulators that delimit the LAS expression domain. To understand the control of LAS expression, the LAS promoter was analyzed. Complementation experiments with deletion constructs demonstrated that at least 820 bp upstream of the ATG and 3547 bp downstream of the ORF are necessary to restore the wildtype phenotype. The regions around -820 to -800 and -810 to -790 were tested for interaction with transcritionfactor proteins. The first sequences interacted with a MYB transcriptionfactor (MYB22; At4g34990) and the second with two members of the TCP gene family (TCP6; At5g23280 and TCP22; At5g08070). Another regulatory element between -2910 and -1447 close to the translation initiation point is necessary, but not sufficient for complementation. The region between +3133 and +3547 in the 3'-region can fulfill its function. A comparable regulation was demonstrated for the tomato Lateral suppressor (Ls) gene, which requires a 5'-region of 1392 bp and a 3'-region of 2633 bp for full complementation of the mutant phenotype. Transcomplementation experiments showed that the Ls downstream region of tomato is also functional in Arabidopsis. These observations lead to the assumption that the same elements in both 3'-regions are present. In addition T-DNA insertion lines were characterized to identify regulatory elements in the promoter region. In one of these insertion lines a lateral suppressor phenotype was found. Next to the insertion at -696 (SALK 50989) a deletion of around 430 bp may be the reason for the las-4-phenotype. Through in silico analysis MADS-BOX protein binding sites were identified. These so called CArG-Boxes are located in the 5'-region of the Arabidopsis LAS gene and in the 3'-region of the tomato Ls ORF. GUS expression studies within the jointless; lateral suppressor double mutant lead to the conclusion that JOINTLESS is important to position the domain for the establishment of the abscissionzone. The signal in ls mutants is less intense than in wt. An autoregulatory pathway could be an explanation for this observation. Another aim of this work was the isolation of LAS homologous genes from barley. Two barley mutants called uniculm1 and uniculm2 show a strong phenotype in sideshoot initiation (Kirby, 1973). Mapping analysis and sequencing of the candidate genes showed that the uniculm phenotypes are not due to a mutation in one of the LAS homologous genes HvLs1 and HvLs2.English
