Abstract

Die vermutlich endodermale Herkunft der Thyroxin-produzierenden Schilddrüsenzellen war bisher nicht eindeutig für Wirbeltiere gezeigt worden. In dieser Arbeit konnte das Endoderm von Zebrafischembryonen durch Injektionsversuche markiert und sein weiteres Entwicklungsschicksal verfolgt werden. Es wurde so festgestellt, dass neben dem Pharynxepithel auch die Schilddrüse vollständig aus markierten Zellen gebildet wird. Transplantationsversuche ergaben, dass die Schilddrüsenvorläuferzellen im Shield-Stadium im Bereich der Pharynxvorläuferzellen zu finden sind. Beide Beobachtungen stehen im Einklang mit der Hypothese, dass sich das Schilddrüsenprimordium als Divertikel vom ventralen Pharynxepithel in der Mittellinie ablöst und somit als Organ endodermaler Abstammung betrachtet werden kann. has mutante Zebrafischembryonen, die über keine funktionale aPKC lambda verfügen, zeigen bisher im Zebrafisch nicht beschriebene Defekte der Relokalisierung. In Transplantationsexperimenten mit tar* exprimierenden und has Morpholino-Oligonukleotiden beinhaltenden Zellen konnte gezeigt werden, dass funktionales aPKC lambda gewebespezifisch im Endoderm benötigt wird, um die vollständige Ablösung des Primordiums aus dem Pharynxepithel zu gewährleisten. Die Beobachtung, dass in Embryonen ohne funktionale aPKC lambda die seitlich verschobenen Primordien von einem stark reduzierten Blutgefäßsystem umgeben sind, ergab die Frage, ob Blutgefäßzellen die Morphologie der Schilddrüse beinflussen können. Durch Transplantation ektopischer Blutgefäßzellen konnte dies bestätigt werden. Untersuchungen zur apiko-basalen Polarität der Schilddrüse zeigen, dass die Zellen des Schilddrüsenprimordiums im Wildtyp zum Zeitpunkt der Ablösung aus dem Pharynxepithel mit der Etablierung der apiko-basalen Polarität beginnen. Sie bilden ein vollkommen polarisiertes Follikelepithel, sobald sie das Pharynxepithel verlassen haben. In dieser Form relokalisiert das Primordium weiter bis zu seiner finalen Position tief ins hypopharyngiale Mesenchym. Der Prozess der Polarisierung und Epithelbildung ist in has Morphants gestört. Es wird kein wildtypisches, einschichtiges Follikelepithel gebildet. Die Lokalisierung von Zellpolaritätsmarkern aber auch die des Schilddrüsenhormonvorläuferproteins Thyroglobulin (TG) ist verändert. Außerdem zeigen has Mutanten und Morphants Defekte während des Differenzierungsprozesses in hormonproduzierende Zellen. Die Analyse der Embryonen mit fehlender aPKC lambda Funktion konnte neue Aspekte der Etablierung von apiko-basaler Polarität der Schilddrüsenzellen aufzeigen und die has Mutation als einen neuen Auslöser für angeborene Hypothyreose identifizieren.

The endodermal origin of thyroid hormone producing cells in the thyroid was not proven in vertebrates. In order to address this question, zebrafish endoderm was marked and the development of these cells was traced. The lineage analysis reveals that thyroid follicle cells are exclusively recruited from the pharyngeal endoderm. Mosaic analysis in tar* grafted host embryos further suggests that the thyroid progenitors are part of the pharyngeal progenitors. Such an origin is later reflected by the evagination of the midline diverticulum from the pharyngeal epithelium. These results show that the thyroid in zebrafish is an endoderm-derived organ. The zebrafish heart and soul (has) mutant lacking aPKC lambda shows defects in this evagination process. Transplantation experiments involving tar* mRNA and has morpholino show that aPKC lambda is tissue-specifically needed within the endoderm for complete evagination of the whole thyroid primordium. Blood vessels surrounding thyroid primordium are dramatically reduced and the thyroid primordium frequently loses its midline orientation in has morphants. This raised the question, if blood vessel cells can influence the morphology of the thyroid. Transplantation of ectopic blood vessel cells confirmed this. Analysis of apico-basal polarity of the relocating thyroid primordium reveals that wildtype cells are starting to polarize at the onset of evagination. Concomitant with the completion of the evagination process thyroid cells are completely polarized and start to form a proper single layered epithelium. They further relocate as epithelium deep into the hypopharyngeal mesenchyme. has morphants show severe defects during epithelia formation. The thyroid in these embryos is not single layered and forms multiple lumina. This is also reflected in altered TG localisation. In addition aPKC lambda deficient embryos frequently show defects in the differentiation process of thyroid hormone producing cells. Analysis of theses embryos reveals new aspects of establishing apico-basal polarity and serves as a new model for defects leading to thyroid hypothroidism.English
