Abstract

Die anterior-posteriore und dorso-ventrale Achse von Drosophila werden bereits früh in der Oogenese durch die Polarisation des Mikrotubuli-Cytoskeletts und der spezifischen Lokalisation der posterioren Determinante osk mRNA, der anterioren Determinante bcd mRNA und dem dorso-ventralen Induktor grk mRNA in der Oocyte determiniert. In einem dieser Arbeit vorausgegangen Screen zur Identifizierung von neuen Genen für die Regulation der Oocytenpolarität wurden die beiden letalen Komplementationsgruppen 32.17/11.229 und 7.460/7.134 entdeckt, die in der vorliegenden Arbeit genetisch und molekular charakterisiert wurden. Die Identifizierung der mutanten Gene erfolgte über meiotische Kartierung mit klassischen Markern und molekularen Markern von einer neu etablierten Polymorphismenkarte für das Chromosom 2R sowie Kartierung mittels neu generierter, molekular definierter Defizienzen. Durch molekulare Identifizierung von Punktmutanten wurde für die Komplementationsgruppe 32.17/11.229 gezeigt, dass sie einem Homologen von thoc5 entspricht. Dies konnte durch Rettungsexperimente bestätigt werden. In thoc5 Mutanten ist der Transport der osk mRNA zum posterioren Pol in den Stadien 9-10 in der Oocyte gestört und die Reduktion der grk mRNA führt zur Ventralisierung der Eikammer. Mit HA-Thoc5 und GFP-Thoc5 Konstrukten wurde gezeigt, dass Thoc5 überwiegend in den Zellkernen vorkommt. Hier konnte Thoc5 als Teil des konservierten THO-Komplexes nachgewiesen werden. Weiterhin stellte sich heraus, dass der THO-Komlex in der Keimbahn mit den beiden Komponenten Mago und Y14 des Exon-Exon-Junction-Komplex interagiert, von denen bekannt ist, dass sie Teil der osk mRNP Transportmaschinerie sind. Die hier erzielten Ergebnisse legen nahe, dass Thoc5 die korrekte Assemblierung der osk mRNP in den Nährzellkernen kontrolliert, was wiederum Voraussetzung für die korrekte Lokalisation der posterioren Determinante osk mRNA zum posterioren Pol im Cytoplasma der Oocyte in den Stadien 9-10 ist. Der Phänotyp der Komplementationsgruppe 7.460/7.134 manifestiert sich ebenfalls in der Misslokalisation der osk mRNA und in der Ventralisierung der Eier. Dabei ist die Polarisation des Mikrotubuli-Cytoskeletts nicht gestört. Das mutante Gen konnte auf eine Region von 20 kb mit vier Genen unbekannter Funktion bei der Oocytenpolarität eingegrenzt werden. Die endgültige Identifizierung des Gens soll mit Rettungsexperimenten erfolgen. Die hier vorliegende Arbeit beschreibt zwei neue Komponenten, die für die Regulation der anterior-posterioren und dorso-ventralen Oocytenpolarität in Drosophila essentiell sind.

The anterior-posterior and dorso-ventral axes of Drosophila are already defined early during oogenesis by the polarisation of the microtubule cytoskeleton and the localisation of the posterior determinant osk mRNA, the anterior determinant bcd mRNA and the dorso-ventral inductor grk mRNA in the oocyte. A previously performed screen for new mutants regulating oocyte polarity identified two lethal complementation groups 32.17/11.229 and 7.460/7.134. This work deals with the genetic and molecular characterisation of these two complementation groups. The mutant genes were identified through meiotic mapping with classical markers and molecular markers from a newly established polymorphism map for chromosome 2R as well as mapping using newly generated, molecular defined deficiencies. The molecular identification of point mutants suggested that the complementation group 32.17/11.229 corresponds to a homolog of thoc5. This could be confirmed by rescue experiments. In thoc5 mutants the transport of the osk mRNA to the posterior pole of the oocyte is blocked during stages 9-10 and the reduction of the grk mRNA leads to the ventralisation of the egg chamber. Using HA-Thoc5 and GFP-Thoc5 constructs Thoc5 is shown to localise predominantly in nuclei, where Thoc5 is part of the conserved THO-complex. The THO-complex is proven to interact, in the germline, with the components of the exon-exon junction complex Mago and Y14, which are both known constituents of the osk mRNPs transport machinery. Our results suggest that Thoc5 controls the assembly of the osk mRNPs in the nurse cell nuclei and that this in turn is a pre-condition for the proper localisation of the posterior determinant osk mRNA at the posterior pole in the cytoplasm of the oocyte during stages 9-10. The phenotype of complementation group 7.460/7.134 manifests itself as a mislocalisation of the osk mRNA and the ventralisation of the eggs without affecting the polarisation of the oocyte microtubule cytoskeleton. The mutant gene could be confined to a region of 20 kb containing four genes with unknown function in oocyte polarity. The final identification of the gene will be done by rescue experiments. This study describes two novel components which are essential for regulating anterior-posterior and dorso-ventral oocyte polarity in Drosophila.English
