Abstract

Das Protein Urim (Up regulated in metastasis) wurde zunächst differentiell exprimiert in metastasierenden und nicht-metastasierenden Tumorzellen gefunden. Weitere Untersuchungen zeigten dessen Homolgie mit dem Protein NLVCF (Nuclear localization signal containing protein deleted in Velo-Cardio-Facial syndrome). Die Funktion und die Bindepartner von Urim waren bisher unbekannt. Durch Ko-Immunopräzipitationen identifizierten wir Hira als einen Interaktionspartner. Mit Hilfe von Hira WD 40-Mutanten zeigten wir, dass Urim an die N-terminale WD 40-Domäne von Hira bindet. Weiterhin sind im Urim-Hira-Komplex die Histone H2B, H3 und H4 als Interaktionspartner gebunden, was eine Beteiligung des Komplexes am Histonmetabolismus und der Chromatinassemblierung wahrscheinlich macht. Der Hira-Urim-Komplex wird zellzyklusphasenabhängig assembliert. Hira, Urim und die Histone H2B, H3 und H4 kolokalisieren in der M-Phase, jedoch nicht in den anderen Phasen des Zellzyklus. Dieses steht in Übereinstimmung mit der Kolokalisation von Hira und Urim im Zytoplasma und Zellkern von M-Phase Zellen, wie es durch Doppelmarkierungen beider Proteine in der Zelle nachgewiesen wurde. In den übrigen Phasen des Zellzyklus liegt zytoplasmatisches Urim unabhängig von im Zellkern lokalisiertem Hira vor. Da Urim und Hira in der S-Phase des Zellzyklus, in dem die replikationsabhängige Chromatinassemblierung erfolgt, nicht interagieren, ist eine Beteiligung des Komplexes an Ereignissen in dieser Zellzyklusphase unwahrscheinlich. Durch die Komplexierung der Proteine Urim und Hira mit Histonen in der M-Phase ohne Bindung des Histon-Chaperons Asf1a liegt eine Beteiligung an der replikationsunabhängigen Chromatinassemblierung nahe. Möglicherweise bilden Hira und Urim einen alternativen Weg der replikationsunabhängigen Chromatinassemblierung, der nicht auf die Bindung von Asf1a an Hira angewiesen ist. Urim hat somit durch Bindung von Hira möglicherweise seine Funktion in der Bereitstellung eines Asf1a unabhängigen Weges der replikationsunabhängigen Chromatinassemblierung.

Initially Urim (Up regulated in metastasis) was developed as a differentially expressed protein in metastasizing and non metastasizing tumors. Further analysis displayed its homology to the protein NLVCF (Nuclear localization signal containing protein deleted in Velo-Cardio-Facial syndrome). Until now the function and binding partners of Urim were unidentified. Using Co-immunoprecipitation assays we identified Hira as an Urim interaction partner. Via analysis of the interaction from Hira WD 40-domain mutant proteins to Urim, we identified the binding domain as the N-terminal Hira WD 40-domain. Anymore we could identify core-histones H2B, H3 and H4 as Urim interactors, indicating that the complex is involved in histone metabolism and chromatin assembly. The Urim-Hira complex assembles in a cell cycle dependent manner. Via Co-immunoprecipitation of the protein Urim we displayed the complex formation of Hira, Urim and the histone proteins H2B, H3 and H4 in M-phase of the cell cycle, but not in the other cell cycle phases. This is consistent with the co-localization of Urim and Hira in M-phase in the cytoplasm and the nucleus as shown here via immunhistochemical double fluorescence staining of both proteins. In all other cell cycle phases cytoplasmatic localized Urim does not interact with nucleus localized Hira. As we could not detect any Hira-Urim interaction in S-phase of the cell cycle, where replication dependend chromatin assembly occurs, we suggest that complexation of Urim and Hira is not necessary for events in this cell cycle phase. The complex formation of Urim, Hira and the histone proteins in M-phase without participation of the Asf1a protein is more likely to show a possible involvement of Urim in the replication independent chromatin assembly. Possibly Urim and Hira establish an alternative way in replication independent chromatin assembly without binding Asf1a. So we conclude that the function of Urim is important due to an alternative Asf1a independent way of replication independent chromatin assembly.English
