Abstract

In E. coli wurde bereits eine Vielzahl von Aminosäureexportern beschrieben. Allerdings liegen zum Export aromatischer Aminosäuren bisher nur wenige Daten vor. Ziel dieser Arbeit war es, den Export von Tryptophan zu charakterisieren und beteiligte Transportproteine zu identifizieren. Dazu kamen biochemische, mikrobiologische und genetischen Methoden zum Einsatz Durch Beladung mit Tryptophan-haltigen Dipeptiden konnte der darauf folgende Export von Tryptophan charakterisiert werden. Die Messung bedingte den Einsatz von Zellen mit delta_tnaAB Hintergrund, um den internen Abbau und die Wiederaufnahme bereits exkretierten Tryptophans zu unterbinden. Es konnte gezeigt werden, dass der Export von Tryptophan ein sekundär aktiver Prozess ist und dass die dafür verantwortlichen Transporter konstitutiv exprimiert werden. Die Identifikation putativer Tryptophanexporter geschah einerseits mittels eines bioinformatischen Ansatzes und gezielter Deletion bzw. Expression von Kandidaten. Eine Alternative stellte die Transformation von E. coli mit Genbanken und die Selektion auf Resistenz gegen das toxische Aminosäureanalogon 5-Methyl-Tryptophan (5MTrp) dar. Die Analyse der Plasmide, die dabei eine Resistenz verliehen, erlaubte die Identifikation putativer Tryptophanexporter. In der Dipeptidfütterung konnte gezeigt werden, dass die Expression von yddG bzw. yedA unter Kontrolle ihrer nativen Promoter zu einer Verringerung der internen Akkumulation von Tryptophan um ca. 50% führte. In Messungen mit radioaktiv markiertem Tryptophan wurde für Expressionsstämme dieser Gene ebenfalls eine gegenüber dem Wildtyp verringerte interne Akkumulation von Tryptophan nachgewiesen. Weiterhin vermitteln die beiden Kandidaten eine erhöhte Resistenz gegen 5MTrp und YddG auch eine Resistenz gegen 3-Fluoro-Phenylalanin. Die Deletion von yddG und yedA bzw. beider Gene führte zu keiner erhöhten Akkumulation von Tryptophan. Es muss also noch mindestens ein weiterer Tryptophanexporter existieren.

A number of amino acid exporters has already been described in E. coli. However, the export of aromatic amino acids has not been studied in detail so far. The aim of this work was to characterise the export of tryptophan and to identify transporters involved in tryptophan excretion. For this purpose biochemical, microbiological and genetic methods have been used. By loading cells with tryptophan containing dipeptides the subsequent export of tryptophan could be characterised. Delta_tnaAB cells were used to exclude internal degradation and re-import of already excreted tryptophan. It could be shown that the export of tryptophan is a secondary active process and that the transporter coding gene is constitutively expressed. The identification of putative tryptophan exporters was done using a bioinformatic approach with deletion and expression of candidates. An alternative approach was the use of E. coli gene banks and the selection of clones that are resistant to the toxic amino acid analog 5-methyl-tryptophan (5MTrp). By analysing plasmids that conferred resistance, the identification of putative tryptophan transporters was possible. Dipeptide feeding assays in cells that expressed yddG and yedA under control oh their native promoters showed a decrease of internal accumulation of tryptophan of about 50% compared to the reference strain. In measurements with radioactive labelled tryptophan it also was possible to demonstrate a reduced level of tryptophan accumulation. Furthermore, both candidates conferred higher resistance towards 5MTrp and yddG expression even increased the resistance towards 3-fluoro-phenylalanine. Deletion of yddG or yedA or both did not lead to a higher accumulation of tryptophan. Hence, there must be at least one as of yet undiscovered exporter of tryptophan on E. coli.English
