Abstract

Natürliche Killer (NK) Zellen sind Lymphozyten des angeborenen Immunsystems und spielen eine wichtige Rolle bei der Erkennung von Tumorzellen und Tumor surveillance. Das intrazelluläre Protein HLA-B assoziierte Transkript 3 (BAT3) ist ein Ligand für den NK Zellrezeptor NKp30. Dieser ist ein Mitglied der natural cytotoxicity receptors (NCRs) und gehört zu den aktivierenden Rezeptoren der NK Zellen. Unty-pisch für NK Zell-Liganden ist, dass BAT3 konstitutiv intrazellulär exprimiert und erst in Antwort auf zellulären Stress in das extrazelluläre Milieu abgegeben wird. Der ex-trazelluläre Faktor interagiert mit NKp30 auf NK Zellen und kann die NK Zell-Funktion beeinflussen. In dieser Arbeit wurde gezeigt, dass BAT3 über einen exosomalen Transportweg aus der Zelle transportiert wird. Exosomen sind extrazelluläre Membranvesikel von 50-100 nm, die eine wichtige Rolle bei der Zell-Zell-Kommunikation spielen. BAT3 konnte auf der Oberfläche von Exosomen nachgewiesen werden. Darüber hinaus korrelierte die Sekretion von BAT3 mit der nSMase2-Aktivität, einem Enzym, das für die Biogenese von Exosomen essentiell ist. Weiterhin zeigten Deletionsmutanten, dass der N-Terminus von BAT3 für den exosomalen Transport absolut notwendig ist. Dieser Sequenzabschnitt diente als "Exosomen Targeting" Sequenz für ein Reporterprotein (GFP). Der Sequenzbereich umfasst eine UBL-Domäne und eine HRS-Bindestelle. Es ist bekannt, dass das HRS-Protein aus dem ESCRT-0-Komplex mit ubiquitinierten Proteinen interagiert und die Verpackung von Proteinen in/auf Exosomen steuert. Co-Immunpräzipitationen zeigten eine Interaktion von BAT3 und HRS. Die Aktivität von NK Zellen ist bei vielen hämatopoetischen Tumoren gehemmt. In dieser Arbeit wurde die Funktion von BAT3 bei der B-CLL untersucht. Das Expres-sionslevel von BAT3 war bei 2/3 der analysierten B-CLL Seren (70) im Vergleich zu gesunden Probanden erhöht. BAT3 lag hier, anders als bei gesunden Spendern, aber nicht exosomal-gebunden vor, sondern fast ausschließlich als lösliches Protein. Die Inkubation von NK Zellen gesunder Spender mit Seren von CLL Patienten oder mit rekombinantem, löslichem BAT3 inhibierte die NK-Zytotoxizität drastisch. Im Gegensatz zu gesunden Spendern (10/10) exprimieren Patienten mit B-CLL (8/10) eine BAT3 Splicevariante, bei der die HRS-Bindesequenz deletiert ist. Dies könnte ein Grund für die defiziente, exosomale BAT3 Expression sein, die letztlich zur verminderten NK Zell-Aktivität führt. Zu Beginn dieser Arbeit gab es noch keine genetischen Modelle zum Studium von NK Zellen. Aus diesem Grund wurde mit der Etablierung eine NK Zell-spezifischen Cre-Mauslinie begonnen. Die Expression von Cre stand dabei unter dem Einfluss des humanen Minimalpromotors von NKp46. Das Konstrukt war in Zelllinien funktionell und ist somit zur Herstellung transgener Linien geeignet. Dieses Tiermodell könnte zur Untersuchung der in vivo Rolle von BAT3/NK-Aktivität für die Pathogenese der B-CLL eingesetzt werden.

Natural Killer (NK) cells are lymphocytes of the innate immune system and play a fundamental role in the defense against tumor cells and their surveillance. The nuc-lear protein HLA-B associated transcript 3 (BAT3) is a ligand for NKp30, which is a member of NCRs (Natural Cytotoxicity Receptor) and belongs to the activating NK cell-receptors. Expression of BAT3 is stress-induced and results in the secretion into the extra-cellular environment. There, it interacts with NKp30 on NK cells and has an influence on NK cell-function. In this work it was proven, that BAT3 is transported out of the cells via an exosomal pathway. Exosomes are extra-cellular vesicles with a size of 50 to 100 nm and play an important role in cell-cell communication. BAT3 was detected on the exosomal surface. Furthermore the secretion of BAT3 correlated with the activity of nSMase2, an enzyme which is essential for the biogenesis of exosomes. In addition, deletion mutants showed that the N-terminus of BAT3 is most important for the exosomal transport. This part of the sequence served as an �exosomal targeting� sequence for a reporter protein (GFP). The sequence range comprises an UBL domain and a HRS binding motif. It is known that the HRS protein from the ESCRT-0 complex interacts with ubiquinated proteins and controls their packing in/on exosomes. Co-immunoprecipitation showed an interaction between BAT3 and HRS. NK cell-activity is inhibited in many hematopoietic tumors. In this work the function of BAT3 regarding B-CLL was investigated. In 2/3 of the analyzed B-CLL sera a higher BAT3 expression level compared to healthy donors was shown. But compared to healthy donors, BAT3 was not exosomal bound � it was almost exclusively a soluble protein. NK cytotoxicity was drastically inhibited, when NK cells of healthy donors were incubated with B-CLL sera or recombinant, soluble BAT3. In contrast to healty probands (10/10), B-CLL patients (8/10) expressed a BAT3 splice variant with a deleted HRS binding domain. This could be the reason for the decreased expression of exosomal BAT3, which results in a reduced NK cell-activity. Beginning this work, no genetic model for the research of NK cells existed. For this reason a NK cell-specific Cre mouse line was developed. The expression of Cre was controled by the human minimal promoter NKp46. This construct was functional in cell lines and is useful for the creation of transgenic lines. These animal models could be used for the analysis of the in vivo role of the BAT3/NK activity in the pathogenesis of B-CLL.English
