Abstract

In dieser Arbeit wird eine effiziente, höchst stereoselektive Synthese von (2R) α Toco¬pherol, dem bedeutensten Vertreter der E Vitamine beschrieben. Als Schlüsselschritt dient eine asymme¬trische konjugierte Addition an ein aktiviertes Chromenon, für das ein verlässliches und potentiell maßstabsvergrößerbares Herstellungsverfahren entwickelt wurde. Die asymmetrische Cu-katalysierte 1,4 Addition von AlMe3 zur Erzeugung des quaternären Stereozentrums erfolgt (unter Verwendung eines MonoPhos®-Liganden) in ausgezeichneter Selektivität und Ausbeute. Weiterhin wird eine Methodik zur Ni-katalysierten 1,4-Addition von verschiedenen Aluminiumorganylen an eine Vielzahl C-2 substituierter Chromenone vorgestellt, die wahrscheinlich über einen Radikal-Mechanismus verläuft.

This work describes an efficient, highly stereoselective synthesis of (2R) α Toco-pherol, the most prominent member of the Vitamin E family. As a keystep serves a conjugate-addition to an activated chromenone substrate, for which a reliable preparation procedure was developed. The asymmetric Cu-catalyzed 1,4-addition of AlMe3 for the generation of the quaternary center (in the presence of a MonoPhos® ligand) afforded the product in excellent yield and selectivity. In addition to this, conjugate addition of several Al organyls to related substrates was shown to proceed under Ni-catalysis, most likely via a radical mechanism.
