Abstract

In jüngster Zeit wurden erfolgreich biotechnologische Verfahren für die industrielle Produktion von Pantothensäure (Vitamin B5) mit Escherichia coli etabliert. Hierbei werden dem Nährmedium hohe Konzentrationen der Vorstufe beta-Alanin (3-Aminopropionsäure) zugesetzt. beta-Alanin wird von den Bakterien aufgenommen und auf biochemischem Wege zu Pantothensäure umgesetzt. Diese wird von den Zellen exkretiert und kann dann aus dem Medium isoliert werden. Da die Pantothensäure-Produktion unmittelbar von den zwei Transportprozessen abhängig ist, können diese die Produktivität limitieren. Die beteiligten Transportsysteme waren jedoch bisher noch nicht bekannt. Das vorrangige Ziel dieser Arbeit war deshalb deren Identifikation und Charakterisierung. Wie gezeigt werden konnte, wird beta-Alanin in einem energieabhängigen Prozess durch den Aminosäurecarrier CycA aufgenommen. Das entsprechende Gen aus E. coli K12 MG1655 konnte kloniert und in verschiedenen Stämmen überexprimiert werden. Hierdurch ließ sich eine fast sechsfache Steigerung der Aufnahmeaktivität erzielen, und die Expression im Produktionsstamm FV5069 führte zur Verdopplung der spezifischen Produktivität. Die Steigerung der beta-Alanin-Aufnahmeaktivität eröffnet somit eventuell neue Möglichkeiten zur weiteren Verbesserung von Produktionsstämmen für Pantothensäure. Für die Identifikation des Pantothensäure-Exkretionscarriers wurde eine Transposonmutanten-Bank etabliert, deren Klone dann auf ihre Fähigkeit zur Exkretion von Pantothensäure untersucht wurden. Der gesuchte Transporter konnte so zwar nicht gefunden werden, jedoch zeigte die Analyse der Mutanten vielfältige Verbindungen zwischen der Pantothensäure-Biosynthese und anderen Stoffwechselwegen auf. Dabei scheint insbesondere die Bildung von 5,10-Methylentetrahydrofolat eine wichtige Rolle zu spielen.

Biotechnological processes for the industrial production of pantothenic acid with Escherichia coli have been developed recently. Herein high concentrations of beta-alanine (3-propionic acid) have to be added to the culture medium. beta-alanine is taken up by the bacteria and biochemically converted to pantothenic acid. The cells excrete the product, which can then be purified from the fermentation broth. Since pantothenic acid production is directly dependent on the two transport processes, their limitation can lower the productivity. Despite of this essential role, the responsible carriers have been unknown so far. Therefore, the predominant goal of this work was their identification and characterization. This thesis revealed that beta-alanine uptake is mediated in an energy-dependent manner by the amino acid carrier CycA. The corresponding gene of E. coli K12 MG1655 has been cloned and overexpressed, leading to a nearly six-fold increase of the beta-alanine uptake rate. Additionally, overexpression in the production strain FV5069 doubled its specific productivity. The enhancement of beta-alanine uptake may therefore be way to further improve pantothenic acid production strains. To identify the carrier responsible for pantothenic acid excretion a set of transposon mutants has been established and the cells were screened for their ability to excrete pantothenic acid. Although the carrier could not be found by this approach the analysis of the mutants revealed many connections of pantothenic acid biosynthesis and other metabolic pathways. Especially the formation of 5,10-methylenetetrahydrofolate seems to play an important role.English
