Abstract

Ziel dieser Arbeit war eine Ausweitung des Verständnisses der genetischen Mechanismen, die die Gastrulationsprozesse in Drosophila melanogaster kontrollieren. Das besondere Interesse galt dabei dem Beginn der Morphogenese der Mesodermanlage, der Ventralfurchenbildung. Dazu wurden mehrere Ansätze verfolgt. Einerseits wurde die Funktion zweier bereits bekannter Faktoren untersucht. Snail und Twist sind zwei Transkriptionsfaktoren, die für die Entwicklung des Mesoderms essentiell sind, deren genaue Funktion für diesen Prozess allerdings nicht geklärt ist und daher näher analysiert werden sollte. Die im Verlauf der Arbeit gesammelten Ergebnisse unterstützen die Annahme, daß der Zinkfinger-Transkriptionsfaktor Snail während der Gastrulation eine notwendige Funktion als Repressor hat. Darüber hinaus wurden Indizien gesammelt, die darauf hinweisen, daß die Rolle von Snail über die eines bloßen Repressors hinausgeht. Untersuchungen der Zielgene des bHLH-Transkriptionsfaktors Twist, seiner Ausfallmutation und damit verbundene Rettungsexperimente legen den Schluß nahe, daß Twist mehr Zielgene haben muß als bisher bekannt sind. Im Einklang damit erfolgte der Versuch, neue genetische Loci zu identifizieren, die für eine normale Mesodermmorphogenese gebraucht werden. Mit Hilfe einer Sammlung von Fliegenstämmen mit autosomalen Deletionen wurde dazu in einem genomweiten Screen nach Genen gesucht, die an der Ventralfurchenbildung im Drosophila-Embryo beteiligt sind. Vier chromosomale Regionen konnten identifiziert werden, deren Anwesenheit für eine normale Gastrulation notwendig ist. Zwei dieser Loci wurden näher untersucht. In einem der beiden Fälle konnte das verantwortliche Gen, tribbles, identifiziert werden. Es ist Teil eines bis dahin unbekannten Mechanismus, der während der Gastrulation von Drosophila für die Koordination von Proliferation und Morphogenese notwendig ist, was den betroffenen Zellen die rasche und ungestörte Abfolge dieser beiden zellbiologisch konträren Prozesse ermöglicht.

The aim of this work was to extend the understanding of the genetic mechanisms which control gastrulation in Drosophila. It focussed on the formation of the ventral furrow which marks the beginning of morphogenesis of the mesodermal anlage. The problem was approached from different directions. On one hand the function of two already known factors was examined. Snail and Twist are two transcription factors which are essential for development of the mesoderm, but it is unclear in which way they control mesoderm morphogenesis. The results of this work support the idea that repression by the zinc finger transcription factor Snail is a major and essential part of its activity. In addition the results suggest that Snail might not only be a repressor but may be also an activator of gene transcription. Analysis of the phenotype of twist mutants as well as rescue experiments employing its known target genes strongly suggest that Twist must have more target genes then are known so far. In parallel to this a genetic screen making use of a collection of fly stocks carrying large autosomal deletions was performed. It aimed at identifying genetic loci, that uncover new genes involved in gastrulation. Four loci could be identified whose presence is necessary for normal formation of the ventral furrow. Two of this loci were examined in more detail. In one of the two cases the corresponding gene, tribbles, could be identified. Analysis of tribbles and its loss of function phenotype showed that it is part of a so far unknown mechanism which coordinates proliferation and morphogenesis of the mesoderm, thereby avoiding conflicting cell biological processes to happen simultaneously within the same cells.English
