Abstract

HGF ("hepatocyte growth factor") ist ein pleiotrophes Zytokin, das in einer mesenchymal-epithelialen Interaktion an den transmembranen Rezeptor MET bindet. Infolgedieser Liganden-Rezeptor-Interaktion werden verschiedene Signalwege aktiviert, die in der Zielzelle sowohl zu einer Stimulierung der Proliferation, Morphogenese, Angiogenese und Migration als auch zu einer Hemmung der Apoptose führen können. Das HGF/MET-Signaltransduktionssystem beeinflußt somit Prozesse der Embryogenese und der Wundheilung verschiedener Organe einschließlich der Haut. In der kutanen Wundheilung sind die spezifischen molekularen Wirkungsmechanismen von HGF in Abgrenzung zu anderen Zytokinen bislang jedoch nicht verstanden. Deshalb wurde in dieser Arbeit der Einfluß von HGF im Vergleich zu anderen Wachstumsfaktoren (KGF und GM-CSF) auf das Expressionsmuster in Keratinozyten während der kutanen Wundheilung analysiert. Dafür wurde das heterologe "feeder-layer" Kokultur-Wundheilungsmodell aus humanen primären Keratinozyten und murinen c-Jun-/- Fibroblasten eingesetzt, da es ermöglicht, die spezifischen Einflüsse dieser Zytokine auf das Expressionsmuster von Keratinozyten unter Ausschluß von sekundären oder kompensatorischen Effekten miteinander zu vergleichen. Mittels "GeneChip® Expression Array"-Analysen wurden sowohl gemeinsame als auch spezifische Zielgene von HGF, KGF und GM-CSF identifiziert und deren Regulation exemplarisch an FRA1, CTGF, TNFAIP3, uPAR und K10 in der semiquantitativen "realtime" PCR bestätigt. Am Beispiel von uPAR konnte nachgewiesen werden, daß sich die zytokinabhängige mRNA-Syntheseregulation auch auf die Proteinbiosynthese auswirkt. Funktionelle Konsequenzen der mRNA-Expressionsregulation von uPAR wurden in vitro ausschließlich bei der HGF-induzierten Migration durch einen Migrationstest mit neutralisierendem uPAR-Antikörper belegt. Darüber hinaus konnte in der murinen Wundheilung in vivo nachgewiesen werden, daß MET und uPAR in aktivierten Keratinozyten insbesondere innerhalb der "migration tongue" bei gleichzeitiger Bioverfügbarkeit von HGF koexprimiert werden. Diese Ergebnisse legen eine maßgebliche und spezifische Funktion von HGF unter der Beteiligung von uPAR bei der Bildung der "migration tongue" nahe. In dieser Arbeit wurde somit ein in vivo nahes System etabliert und verifiziert, mit dessen Hilfe der spezifische Einfluß von HGF und anderen wundheilungsrelevanten Zytokinen auf die Expression von Zielgenen in primären Keratinozyten analysiert werden kann. Weiterhin konnte am Beispiel von uPAR gezeigt werden, daß die Expression der identifizierten Zielgene von zentraler Bedeutung für das Verständnis der Wirkung von HGF in der kutanen Wundheilung ist.

HGF (hepatocyte growth factor) is a pleiotrophic cytokine that activates its receptor MET in a mesenchymal-epithelial interaction, leading to a variety of cellular responses such as proliferation, migration, morphogenesis, angiogenesis and anti-apoptosis. By these effects the HGF/MET signal transduction system aligns cellular activities during cutaneous wound healing, whereas the molecular mechanisms in comparison to other cytokines are not completely understood. Therefore the impact of HGF versus KGF and GM-CSF on the gene regulation of primary keratinocytes during cutaneous wound healing has been analysed in a model based on a heterologous feeder-layer coculture. Thus the primary influences of these growth factors could be evaluated while their secondary and compensatory effects were omitted. Common and specific target genes of HGF, KGF and GM-CSF have been identified by oligonucleotide array analysis. The regulation of these genes could be exemplarily confirmed for FRA1, CTGF, TNFAIP3, K10 and uPAR by qPCR, and for the latter also by Western Immunoblot. Further, HGF-specific, functional consequences of the uPAR upregulation were proven in vitro by a scratch test with a neutralising uPAR-antibody. In fact, the coexpression of MET and uPAR in activated keratinocytes particularly within the migration tongue as well as the bioavailability of HGF could be detected in vivo during wound healing, indicating both that HGF has a major and specific impact on the formation of the migration tongue and that uPAR is involved in this process. Therefore a wound healing model close to the in vivo situation could be established and verified within this thesis, which enables the analysis of specific influences of HGF and other significant cytokines in wound healing on the expression of target genes in primary keratinocytes. The expression of the identified target genes bears a pivotal relevancy for the understanding of the HGF function within cutanous wound healing.English
