Abstract
The phthalide natural product primarolide A and the isoindolinone primarolide B were isolated from a marine fungus. We hypothesized the biological origin of both primarolides from an ortho-formyl benzophenone precursor, we termed putative pro-primarolide. According to the proposed structures in the literature, a highly variable synthesis strategy was developed, which forms the structural motif of the central benzophenone unit by means of an anionic homo-Fries rearrangement providing the putative pro-primarolide. Furthermore, a conversion into the corresponding phthalide and isoindolinone was realized. However, analysis of the putative synthetic primarolides revealed a structural discrepancy to the primarolides isolated from nature. The constitution of the primarolides was therefore revised and a different substitution pattern based on the natural product massarinin A devised. The synthesis of massarinin A was realized with only minor adjustments of the previously developed strategy. Synthetic massarinin A, was converted into the primarolides A and B by means of a Lewis acid or nucleophile-induced cyclo-isomerization or reaction with aniline, respectively. During the synthesis of primarolide A, a 1,4-aryl shift was observed, which could be followed by means of 13C labeling experiments. Furthermore, the structures of the putative and the real primarolides, as well as those of massarinin A were verified by means of single-crystal structure analysis.
