Abstract

Die Positronen-Emissions-Tomographie (PET) ist zu einem unverzichtbaren Bestandteil in der klinischen Forschung und Diagnostik geworden, da sie die in vivo Beobachtung physiologischer Prozesse auf molekularer Ebene ermöglicht. Eine Voraussetzung für die Durchführung klinischer PET-Messungen ist die Entwicklung und Produktion spezifischer Radiotracer, die im Körper selektiv mit Targetstrukturen wie Tumoren oder Rezeptorsystemen wechselwirken. In der PET ist 18F das am häufigsten eingesetzte Radionuklid. Die direkte Markierung mit [18F]Fluorid erfordert jedoch harsche Reaktionsbedingungen und ist oft nicht für empfindliche Biomoleküle geeignet. In diesem Fall stellen indirekte Methoden unter Verwendung von radiomarkierten Bausteinen eine gute, wenn auch zeitaufwändige und daher weniger effiziente Alternative dar. Ziel dieser Arbeit war die Weiterentwicklung und praktische Anwendung von neuen Methoden zur indirekten Radiomarkierung von Peptiden. Wichtige Zielmoleküle, deren Markierung im Rahmen dieser Arbeit etabliert werden sollte, waren PSMA-selektive Liganden und argininreiche, vollständig D-enantiomere Peptide (D3 und RD2). Ersteres wird für die Detektion des Prostatakarzinoms eingesetzt und das Potential der beiden letzteren Peptide zur ursächlichen Behandlung der Alzheimer-Krankheit wird aktuell untersucht. In dieser Arbeit wurde die Synthese von zwei verschiedenen Markierungsbausteinen und deren Möglichkeiten zur Kopplung an Biomoleküle evaluiert. Zur Konjugation des vielversprechenden Synthons (Z)-2 [18F]Fluorhex 1 en-1-yl(phenyl)iodoniumtosylat ([18F]FHexI+) wurden zunächst verschiedene palladiumkatalysierte Kreuzkupplungsreaktionen mit strukturell einfachen Modellverbindungen untersucht. Nach der Radiosynthese von [18F]FHexI+ im wässrigen Milieu konnten im Anschluss daran sowohl die Sonogashira-Kupplung als auch die Suzuki-Kupplung als Eintopfreaktion in dem Medium durchgeführt werden, in dem auch die Herstellung von [18F]FHexI+ stattfand. Da die Sonogashira-Kupplung die besten Ergebnisse lieferte, wurde die Methode anschließend schrittweise anhand aminosäurebasierter Modellverbindungen wie beispielsweise alkinmodifizierter Lysinderivate evaluiert und weiter optimiert, wobei für die Konjugation mit [18F]FHexI+ RCA von 38 – 88 % erhalten wurden. Die Methode lieferte schließlich in einer Eintopfsynthese das 18F-markierte Konjugat eines Glu-C(O)-Lys basierten PSMA-Liganden in einer isolierten radiochemischen Gesamtausbeute von 35 %. Auch konnte ein radiofluoriertes RD2-Derivat nach HPLC-Reinigung des [18F]FHexI+-Synthons in einer isolierten radiochemischen Gesamtausbeute von ca. 10 % erhalten werden. Beide Tracer wiesen allerdings eine ausgeprägte in vivo Defluorierung auf, so dass sich beide Radiokonjugate nicht für PET Untersuchungen eigneten. Daher wurde im Weiteren als alternatives Synthon 2-[18F]Fluor-5-iodpyridin ([18F]FIPy) zur Konjugation evaluiert. Der Synthesebaustein wurde durch nukleophile Substitution eines Ammoniumtriflat-Vorläufers gefolgt von einer Festphasenextraktion (SPE) schnell und in ca. 80 % RCA erhalten. Die folgende Pd-katalysierte chemoselektive S-Arylierung wurde im Hinblick auf eine möglichst geringe Vorläufermenge optimiert. Aufgrund der unzureichenden chemischen Reinheit nach der SPE musste [18F]FIPy zur Markierung des all-D Peptids zunächst mittels HPLC gereinigt werden, so dass das Markierungsprodukt schließlich in ca. 40 % isolierter Gesamt-RCA erhalten werden konnte. Bei Verwendung höherer Vorläufermengen wurden jedoch selbst nach einer einfachen SPE-Reinigung des Synthons in einem leicht zu automatisierenden Prozess hohe isolierte Gesamt-RCA von ca. 55 % sowohl für einen Glu-C(O)-Cys-basierten PSMA-Liganden als auch für ein D3-Derivat erhalten. Die Tracer waren in vivo stabil und werden gegenwärtig präklinisch evaluiert.

Positron emission tomography (PET) is an indispensable tool in clinical research and diagnostics for the visualization of physiological and pathophysiological processes at the molecular level in vivo. For different clinical examinations, PET requires radiolabeled, tailor-made molecules, so called radiotracers, specifically targeting selected molecular structures. The development and efficient production of these tracers is therefore an essential task in PET research. Direct radiolabeling approaches with 18F, one of the most suitable radionuclides for PET, require harsh reaction conditions and are often not applicable for sensitive biomolecules. However, indirect methods using radiolabeled building blocks are well suited as alternatives although they are more time consuming due to multi step syntheses and thus less effective. The aim of this work was the further development and application of novel indirect radiolabeling methods for peptides. Accordingly, two different peptides have been used for radiolabeling. Radiolabeled PSMA-selective ligands and arginine-rich, fully D-enantiomeric peptides (D3 and RD2) should be prepared in this work. PSMA-selective peptidomimetics are currently being used for the detection of prostate carcinoma and the D-enantiomeric peptides in Alzheimer's research since they potentially enable a causal treatment of this disease which is otherwise not possible so far. For the conjugation of the promising synthon (Z)-2-[18F]fluorohex-1-en-1-yl(phenyl)iodonium tosylate ([18F]FHexI+), various Pd-catalyzed cross-coupling reactions were first investigated using structurally simple coupling partners. After radiosynthesis of [18F]FHexI+ under aqueous conditions starting from [18F]fluoride, subsequently both, the Sonogashira coupling and the Suzuki coupling reaction were carried out as a one-pot reaction directly in the initial labelling mixture. As the Sonogashira coupling provided the highest yields, this conjugation reaction was then evaluated and optimized step by step using amino acid-based model compounds, resulting in RCYs of 38 – 88 % for the coupling with [18F]FHexI+. The method finally afforded the Glu C(O) Lys based PSMA ligand in a one-pot synthesis and, after HPLC purification of the synthon, a RD2-derived peptide in a RCY of approx. 35 % and in an isolated total RCY of approx. 10 %, respectively. However, both tracers exhibited a high in vivo defluorination rate making them unsuitable as PET tracers. Next, 2-[18F]fluoro-5-iodopyridine ([18F]FIPy) was investigated as an alternative synthon. This radiolabeled building block was rapidly prepared in high RCY by nucleophilic substitution of an ammoniumtriflate precursor and purification by solid phase extraction (SPE). The subsequent Pd-catalyzed S-arylation was optimized with respect to the precursor amount. Due to an insufficient chemical purity, low concentrations of all-D peptides could only be labeled with 18F after HPLC purification of the labeling building block [18F]FIPy in an isolated total RCY of approx. 40 %. However, using a higher amount of peptide precursor, even after straightforward SPE purification of the synthon, a high total isolated RCY of approximately 55 % was obtained for both, the Glu-C(O)-Cys based PSMA ligand and the D3 derivative. Additionally, this process is amenable to automation. The tracers were stable in vivo and are currently under evaluation in preclinical studies.English
