Abstract

Im Rahmen dieser Arbeit wurden Expressionsort und -form des Testican-2-Proteins in der Maus mit besonderer Fokussierung auf die Lokalisation im Verlauf der Entwicklung untersucht. Des Weiteren wurde das Verhalten von Testican-2 in Bezug auf Zelladhäsion geprüft und potentielle Bindungspartner identifiziert. Ein polyklonaler Testican-2-Antikörper wurde zu Beginn affinitätsgereinigt und eine verbliebende Kreuzreaktivität gegen eine unbekanntes intrazelluläres Protein erfolgreich abgereichert. Während der embryonalen Entwicklung konnte Testican-2 zum ersten Mal an Embryonaltag (E) 6,5 beobachtet werden. Zunächst lag es assoziiert an die Basalmembran zwischen den Keimblättern vor (E 6,5-9,5), danach zeigte es eine eher Zell-assoziierte Färbung in Herzmyokard (ab E 9,5), Nervensystem und den darin enthaltenen Blutgefäßen (ab E 10,5), Skelettmuskulatur (ab E 14,5), endokrinen Drüsen (ab E 16,5) und Lunge (ab Postnataltag 1,5). In der adulten Maus war Testican-2 verbreitet im Nervengewebe inklusive Blutgefäßen und in definierten Strukturen endokriner Organe sowie in Lunge zu detektieren, zeigte aber keine Expression mehr in Herz und Muskel. Die Färbung im zentralen Nervensystem war, abgesehen von den Endothelzellen der Blutgefäße, auf Neurone begrenzt. Bei der Analyse der Glykoformen von Testican-2 konnten sowohl GAG-Ketten als auch N-Glykane nachgewiesen werden, wobei rekombinantes Testican-2 Chondroitinsulfat- und Heparansulfat-Ketten aufwies, während bei endogenem Testican-2 nur Heparansulfat-Ketten zu beobachten waren. In Zellbindungsstudien konnte eine schwache, Ionen-abhängige Bindung von Fibroblasten und Keratinozyten an Testican-2 nachgewiesen werden. Mittels Zell-Inhibitionsstudien mit einem anti-b1-Integrin-Antikörper sowie Protein-Protein-Interaktionsstudien war es möglich, a1b1-Integrin als Bindungspartner von Testican-2 zu identifizieren. Die Bindung an a1b1-Integrin war Konformations-abhängig und mit Antikörpern gegen a1- und b1-Integrin signifikant inhibierbar. Anhäsion von RuGli-Gliom-Zellen an Testican-2 zeigte die physiologische Relevanz der Interaktion von Testican-2 und a1b1-Integrin. Dabei war eine Spreitung der Zellen und die Konzentration von a1b1-Integrin an fokalen Kontakten zu beobachten.

In this thesis, the distribution and structure of testican-2 protein with focus on localisation during embryonic development of the mouse have been studied. In addition the properties of testican-2 in cell adhesion assays were analysed and potential binding partners were identified. A polyclonale antibody was raised in rabbit, purified and successfully relieved of a cross reactivity against an unknown intracellular protein. In embryonic development testican-2 was first recognized at embryonic day (E) 6.5, where it was associated with the basement membrane between the germ layers (E 6.5-9.5). Later in the development a more cell-associated staining was seen in heart myocardium (from E 9.5), nervous system including the developing blood vessels (from E 10.5), skeletal muscle (from E 14.5), endocrinal glands (from E 16.5) and lung (from postnatal day 1.5). In adult mouse, testican-2 was expressed in the nervous system including blood vessels, in defined structures of endocrinal glands and in lung, but not in heart and muscle. Except for endothelia cells of blood vessels only neurons were stained in the nervous system. Analysis of the glycostructure revealed that native testican-2 is a heparan sulfate proteoglycan, whereas recombinant testican-2 bears chondroitin and heparan sulfate chains. The attachment of N-glycans to both testican-2 forms could also be proven. In cell binding studies cation-dependent adhesion and spreading of fibroblasts and keratinocytes on testican-2 could be shown. By inhibition of the cell adhesion with an anti-b1-integrin-antibody in combination with protein-protein interaction studies, a1b1 could be identified as a binding partner of testican-2. Binding of the a1b1-integrin was conformation-dependent and could be significantly inhibited by incubation with antibodies against a1- and b1-integrin. Attachment of RuGli-glioma cells to testican-2 revealed the physiological relevance of the interaction between testican-2 and a1b1-integrin. Thereby cell spreading and concentration of a1b1-Integrin in focal contacts could be observed.English
